摘  要:目的评价缺血后处理对大鼠肾缺血再灌注时c-fos蛋白表达的影响。方法成年雄性sD大鼠75只,8~12周龄,体重200~250g,采用随机数字表法,将其分为3组(n=25):假手术组(S组)、缺血再灌注组(I/R组)和缺血后处理组(IPO组)。I/R组和IPO组采用结扎右侧肾蒂,夹闭左侧肾蒂1h,再灌注24h的方法制备大鼠肾缺血再灌注损伤模型。分别于再灌注1、3、6、12和24h时,处死5只大鼠,分离左肾组织,光镜下观察病理学结果,采用免疫组化法检测c-fos蛋白表达。结果与S组比较,I/R组和IPO组再灌注各时点肾组织c-fos蛋白表达上调(P〈0.05),病理学损伤明显;与I/R组比较,IPO组再灌注各时点肾组织c-fos蛋白表达下调(P〈0.05),病理学损伤减轻。结论缺血后处理减轻大鼠肾缺血再灌注损伤的机制与下调肾组织c-fos蛋白表达有关。Objective TO evaluate the effects of isehemic postconditioning (IPO) on c-fos protein expression during renal ischemia-reperfusion (I/R) in rats. Methods Seventy-five adult male Spragne-Dawley rats, aged 8-12 weeks, weighing 200-250 g, were randomly allocated into 3 groups ( n = 25 each) using a random number table: sham operation group (group S), I/R group and IPO group. Renal I/R injury was induced by damping the bilateral renal pedicles for 1 h followed by 24 h of reperfusion in I/R and IPO groups. Five animals were sacrificed at 1, 3, 6, 12 and 24 h of reperfusion and the left kidneys were removed for microscopic examination and for determination of the expression of c-fos protein by immunohistochemistry. Results Compared with S group, the expression of c-fos protein was significantly up-regulated at each time point during reperfusion in I/R and IPO groups. Compared with I/R group, the expression of c-fos protein was significantly down-regulated at each time point during reperfusiou in group IPO. The pathologic changes were significantly attenuated in group IPO as compared with group I/R. Conclusion The mechanism by which IPO attenuates renal I/R injury is related to down-regnlatiou of c-fos protein expression in the renal tissues of rats.
