摘　　要：目的观察CUL4B基因在结肠癌组织中的表达,以及对人结肠癌细胞株HCT116增殖能力的影响。方法运用实时定量荧光PCR(Real-time PCR)技术检测40例结肠癌及配对正常结肠黏膜中CUL4B mRNA的表达量,比较两者表达差异。构建pEGFP-N1-CUL4B真核表达载体,并通过脂质体质粒转染HCT116细胞,运用体外细胞毒性试验(MTT)法研究CUL4B对结肠癌HCT116细胞增殖功能的影响。结果 40例结肠癌组织中CUL4B mRNA表达水平ΔCt为13.21±1.33,对应正常组织表达水平ΔCt为18.61±1.86,结肠癌组织中CUL4B基因表达量明显高于配对正常结肠黏膜组织(P<0.05)。MTT实验结果显示CUL4B可显著促进结肠癌HCT116细胞的增殖,HCT116-GFP-RKO组与阴性对照组、亲本细胞组对比较差异有统计学意义(P<0.05)。结论 UL4B在结肠癌组织中异常高表达,并且可促进结肠癌HCT116细胞株的增殖功能,可能在结肠癌发生发展中起重要作用。Objective To evaluate the expression pattern of CUL4 B in colon cancer and analyze its effect on the proliferation of HCT116 cell lines. Methods The real-time PCR was conducted to quantify the expression of CUL4 B mRNA in the cancerous tissues and the paired normal mucosa from forty patients of colon cancer. The eukaryotic vector of CUL4 B was constructed. The recombinant was transfected into HCT116 cells. The influence of CUL4 B up-regulation on proliferation of colon cancer cells was analyzed using MTT.Results In forty patients with colon cancer,the global expression（ ΔCt） of CUL4 B was 13. 21 ± 1. 33 in tumor tissue and 18. 61 ± 1. 86 in paired normal mucosa. CUL4 B mRNA expression in tumors was significantly higher than that in the normal mucosa（ P〈0. 05）. The results of MTT test showed that cell counts of experimental group were higher than those in the control group. The HCT116 cells proliferation was promoted significantly by up-regulation of CUL4B（ P〈0. 05）. Conclusion CUL4 B gene possess a higher expression level in colon cancer,and up-regulation of CUL4 B can enhance proliferation of HCT116 cells,which indicated that the CUL4 B probably play an important role in the progression of colon cancer.
