摘 要:目的:探讨宫颈癌组织中E-钙粘素(E-Cad)基因5'端CpG岛甲基化与蛋白表达的关系。方法:采用甲基化特异性聚合酶链反应(MS-PCR)和免疫组织化学法分别检测10例正常宫颈鳞状上皮、31例宫颈癌组织中E-Cad基因5'端CpG岛的甲基化状况与蛋白表达情况,并分析其与宫颈癌临床病理特征的关系。结果:1正常宫颈鳞状上皮组织中未发现存在E-Cad基因甲基化状态,而在宫颈癌组织中E-Cad基因的甲基化率为54.84%(17/31);2宫颈癌组织中存在E-Cad蛋白表达降低或缺失,阳性率仅为38.71%(12/31),显著低于正常宫颈组织的100.00%(10/10),差异有统计学意义(P<0.05)。结论:E-Cad基因5'端CpG岛甲基化是宫颈癌中该基因失活的机制之一,可能与宫颈癌的发生、发展有关。Objective : To explore the relationship between methylation of 5′- CpG island of E - cadherin gene and protein expression in cervical cancer. Methods: Methylation specific polymerase chain reaction (MS -PCR) and immunohistochemical method were used to detect methylation condition of 5′- CpG island of E - cadherin gene and protein expression in 10 cases with normal cervical squamous epithelium and 31 cases with cervical cancer, respectively; the relationship with clinicopathological characteristics of cervical cancer was analyzed. Results: There was no methylation of E - cadherin gene in normal cervical squamous epithelium, while in cervical cancer tissue, the methylation rate of E - cadherin gene was 54. 8% ( 17/31 ) ; there was down - regulation or deletion of E - cadherin protein expression in cervical cancer tissue, the positive rate was only 38.71% ( 12/31 ), which was statistically significantly lower than that in normal cervical squamous epithelium ( 100. 00% , 10/10) ( P 〈 0.05 ) . Conclusion : Methylation of 5′- CpG island of E - cadherin gene is one of mechanisms of E - cadherin gene inactivation in cervical cancer, which may be correlated with occurrence and development of cervical cancer.
