摘 要:目的分析山蜡梅叶石油醚萃取物的成分及其对乙酰胆碱酯酶的抑制活性。方法采用乙醇浸提法、液液萃取法得到山蜡梅叶石油醚萃取物,用气质联用法对其进行组成分析并用面积归一化法测定各化学成分的相对百分含量。使用改进了的Ellman比色法检测山蜡梅叶石油醚萃取物对乙酰胆碱酯酶的抑制活性。结果从山蜡梅叶石油醚萃取物中共检出47个化学成分,占其总组成总量的86.456%,其主要成分是9,12,15-十八碳三烯酸乙酯(27.370%);棕榈酸乙酯(17.340%);植醇(11.616%);9,12-十八碳二烯酸乙酯(9.089%)。山蜡梅叶的石油醚萃取物对乙酰胆碱酯酶的抑制活性较弱,且呈剂量依赖关系。结论该研究为山蜡梅叶的综合开发利用提供了科学依据。Objective To analyze the chemical constituents of petroleum ether extracts from leaves of Chimonanthus nitens and study its Aeetyleholinesterase (AChE) inhibitory activity. Methods The petroleum ether extracts from leaves of Chimonanthus nitens were identified by using gas chromatography - mass spectrometry ( GC - MS) , and its AChE inhibitory activity were evaluated with the improved Ellman eolorimetry. Results 47 compounds were separated and identified which accounted for 86. 456% of total extracts from the petroleum ether extracts. The main chemical constituents were Ethyl 9, 12, 15 - octadecatrienoate (27. 370% ), Hexadeeanoie acid, ethyl ester( 17. 340% ), Phytol ( 11. 616% ) ,9,12 - Octadecadienoic acid ethyl(9. 089% ). The inhibition rate of the extracts to AChE activity was low, and higher concentration of the extracts showed higher inhibition rate. Conclusion This study provides scientific foundations for employments and utilization of the leaves of Chimonanthus nitens.
