摘 要:目的观察β-咔啉类生物碱诱导人胃癌细胞株SGC-7901细胞凋亡的形态学改变,探讨其作用机制。方法将β-咔啉类生物碱设定为浓度0、5、10、20、40μg/mL,分别用其处理人胃癌细胞株SGC-7901 48h,透射电镜观察其对SGC-7901细胞超微结构的影响;荧光显微镜观察其对SGC-7901细胞凋亡形态的影响。结果透射电镜结果可见SGC-7901细胞超微结构改变,提示其可诱导细胞凋亡;Hoechst 33258荧光染色分析,在不同浓度β-咔啉类生物碱(0、5、10、20、40μg/mL)作用下,SGC-7901细胞凋亡率分别为(3.25±0.25)%、(6.30±0.39)%、(9.34±0.54)%、(11.33±0.37)%、(19.86±0.55)%,0μg/mL组分别与其他药物浓度组比较,差异具有统计学意义(P<0.05)。结论β-咔啉类生物碱可诱导人胃癌SGC-7901细胞株发生凋亡。Objective To investigate the morphologic influence ofβ-carboline alkaloids on apoptosis,as well as the related inhibiting mechanism,in human gastric cancer cell cell SGC-7901.Methods Human gastric cancer Cell SGC-7901 was pretreated with 5different concentrations ofβ-carboline alkaloids(0,5,10,20,and 40μg/mL).Ultra structure of cell SCG-7901 was observed by transmission electronic microscope,and apoptosis of cell SCG-7901 was observed by fluorescence microscope.Results After 48 htreatment with different levels ofβ-carboline alkaloids,the ultra structure of cell SCG-7901 was changed as showed under transmission electronic microscope.The result of Hoechst 33258 fluorescent staining analysis showed the apoptosis rate was(3.25±0.25)%,(6.30±0.39)%,(9.34±0.54)%,(11.33±0.37)%,and(19.86±0.55)%respectively.There were significant differences in apoptosis rate between concentration levels of 0μg/mL and 5,10,20,and 40μg/mL(P〈0.05).Conclusion β-carboline alkaloids have potential to induce apoptosis of human gastric cancer cell SCG-7901.
