摘  要:目的通过测定卵巢癌组织核因子-κB(NF-κB)与血管内皮生长因子(VEGF)及IL-2的表达,探讨NF-κB通道的激活及其与肿瘤血管形成的关系。方法收集本院2008年~2014年卵巢癌患者47例,Ⅰ期组12例,Ⅱ期组20例,Ⅲ/Ⅳ期组15例,选择正常卵巢20例为对照组。采用免疫组织化学方法(SABC)测定NF-κB、VEGF和IL-2的表达,并用SPSS 19.0软件对数据进行统计学分析。结果 1)对照组可见少量NF-κB、VEGF和IL-2蛋白的表达;与对照组比较,Ⅰ期组、Ⅱ期组和Ⅲ/Ⅳ期组NF-κB、VEGF和IL-2表达均增多(P〈0.05);与Ⅰ期组比较,Ⅱ期组和Ⅲ/Ⅳ期组NF-κB、VEGF表达均增多(P〈0.05),IL-2表达均减少(P〈0.05);与Ⅱ期组比较,Ⅲ/Ⅳ期组NF-κB、VEGF表达增多(P〈0.05),IL-2表达均减少(P〈0.05)。2)Ⅰ期组、Ⅱ期组和Ⅲ/Ⅳ期组NF-κB与VEGF呈正相关(P〈0.05),与IL-2呈负相关(P〈0.05)。结论卵巢癌相关蛋白NF-κB可通过上调VEGF的表达及下调IL-2的表达共同调节肿瘤血管的生成。Objective By measuring expression of nuclear factor kappa B(NF-κB)and vascular endothelial growth factor(VEGF)expression and IL-2in ovarian cancer tissue,to investigate the relationship between the activation of NF-κB pathway and tumor angiogenesis.Methods Choose 47 cases patients with ovarian carcinoma in hospital from 2008 to 2014,12 cases in stageⅠ group,20 cases in stage II,15 cases in Ⅲ/Ⅳ group,20 normal ovarian as control group.Expression of NF-κB,VEGF and IL-2was measured by immunohistochemical method,the data were analyzed by SPSS 19.0software.Results 1)Expression of NF-κB,VEGF and IL-2protein were found a small amount in the control group.Compared with the control group,NF-κB,VEGF and IL-2protein were increased in groupⅠ,ⅡandⅢ/Ⅳgroup(P〈0.05).Compared withⅡ,NF-κB and VEGF were increased,but IL-2protein was decreased in Ⅲ / Ⅳ group(P〈0.05).2)In stage Ⅰ,Ⅱ and group stage Ⅲ/Ⅳ group of NF-κB was positively correlated with VEGF(P〈0.05),and negatively correlated with IL-2(P〈0.05).Conclusion NF-κB signal pathway was activated in tissue with ovarian cancer,that may be associated with up regulating the expression of VEGF and down regulating the expression of IL-2to regulate of tumor angiogenesis.
