摘 要:目的探讨甜菜碱对高脂饲料诱导的非酒精性脂肪肝(non-alcoholic fatty liver disease,NAFLD)小鼠肝脏甘油三酯(triglyceride,TG)水平、病理组织学形态和脂代谢相关基因表达水平的影响。方法将72只雄性C57BL/6小鼠随机分为对照组,NAFLD模型组和1%甜菜碱干预组,每组24只。对照组饲以普通饲料,模型组和干预组均饲以高脂饲料。对照组和模型组均给予普通的饮用水,干预组给予含1%甜菜碱的饮用水。干预12w和18w后,试剂盒测定肝脏TG浓度;肝组织苏木素和伊红(hematoxylin-eosin,HE)染色观察组织形态学变化;反转录聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)检测肝组织脂肪酸合成酶(fatty acid synthetase,FAS)、乙酰辅酶A羧化酶(acetyl-CoA carboxylase,ACC)、脂肪分化相关蛋白(adipose differentiation-related protein,ADRP)、过氧化酶体增殖物激活α受体(peroxisome proliferator-activated receptorsα,PPARα)和微粒体甘油三酯转运蛋白(microsomal triglyceride transfer protein,MTTP)mRNA表达水平;Western blot检测肝组织MTTP蛋白表达水平。结果干预12w后,与对照组相比,模型组小鼠肝脏TG水平明显上升(P<0.01),PPARα和MTTP mRNA表达水平明显下降(P<0.01);与模型组相比,1%甜菜碱干预组小鼠肝脏TG水平、ACC和ADRP mRNA表达水平显著下降(均为P<0.01),及PPARα和MTTP mRNA表达水平呈上升趋势(P>0.05)。干预18w后,与对照组相比,模型组小鼠肝脏TG水平、FAS、ACC和PPARαmRNA表达水平差异无统计学意义,ADRP和MTTP mRNA表达水平明显下降(均为P>0.05);与模型组相比,1%甜菜碱干预组小鼠肝脏TG水平明显下降(P<0.01),PPARα和MTTP mRNA表达水平明显增加(均为P<0.01)。HE染色结果显示,干预组脂肪变性较模型组明显减轻。结论甜菜碱通过调节肝脏脂代谢相关基因的表达,影响TG水平,对高脂饮食诱导NAFLD小鼠具有肝损伤保护作用。Objective To investigate the effects ofbetaine (B) on serum and liver triglyceride (TG), liver histopatho- logic morphology and expression of liPid metabolism-related genes in mice with non-alcoholic fatty liver disease (NAFLD). Methods Male C57BL/6 mice (n=72) were divided randomly into control diet (CD) group, high fat diet (HFD) group, and HFD+B group. The CD group was fed an ordinary diet, while HFD and HFD+B groups fed a HFD. The CD and HFD groups were given Ordinary drinking water, while HFD+B group given drinking water supplemented with 1% betaine (wt/vol). At the end of 12th and 18th week, half of the mice in every group was sacficed, respectively and serum TG was measured. The liver histopatholigical changes were observed by hematoxylin-eosin (HE) staining. The mRNA expressions of fatty acid synthetase (FAS), aeetyl-CoA carboxylase (ACC), adipose differentiation-related protein (ADRP), peroxisome proliferator, activated receptors α (PPARα) and microsomal triglyceride transfer protein (MTTP) in live were measured by reverse transcription-polymerase chain reaction (RT-PCR). Liver MTTP protein was measured by Western blot. Results After 12 w, compared with those in CD group, the level of liver TG was significantly higher, while the mRNA expressions of PPARct and MTTP significantly lower (all P〈 0.01) in HFD group. Compared with those in HFD group, the TG level and the mRNA expressions of ACC and ADRP in HFD +B group were significantly decreased (all P〈0.01), while the mRNA exloressions ofPPARα and MTTP showed an increasing trend (P〉0.05). After 18 w, the TG level and the mRNA expressions of FAS, ACC and PPARα in HFD group were significantly different compared with CD group, while the mRNA expressions of ADRP and MTTP significantly decreased (all P 〈 0.05).
