阿霉素诱导G-四联体形成对SW480细胞端粒酶活性的影响

摘  要:目的探讨G-四联体结构的生物学功能及其阿霉素诱导鸟嘌呤四联体形成对SW480细胞端粒酶活性的影响。方法通过MTT法测定不同浓度阿霉素(0,2.5,5,10,20μg/ml)对SW480细胞的增殖抑制率;通过PCR-TRAP-ELISA检测不同浓度阿霉素对大肠癌SW480细胞端粒酶活性的影响;通过非变性聚丙烯酰胺凝胶电泳迁移实验分析阿霉素对鸟嘌呤四联体结构及其相似物的影响。结果阿霉素抑制SW480细胞的增殖,且随阿霉素浓度升高和作用时间延长,抑制作用逐步增强;阿霉素降低SW480细胞端粒酶活性;阿霉素可使端粒序列重复4次及以上的含GGG串联区的线性核苷酸结构发生改变,形成紧凑的二级结构,导致迁移速度加快。结论阿霉素抑制SW480细胞的增殖,降低其端粒酶活性,其机制可能与阿霉素诱导G-四联体形成有关。Objective To detect the biological function of guanine-quadruplexes structure and its effects on telomerase activity of SW480 cells.Methods The inhibition rates of SW480 cell proliferation after cultured with different concentrations of adriamycin (0,2.5,5,10,20 μg/ml) were measured using MTT assay.The telomerase activities of SW480 cells cultured with different concentrations of adriamycin were detected by PCR-TRAP-ELISA.The effects of adriamycin on structure of guanine-quadruplexes and its analogues were observed by the non-denaturing polyacrylamide gel electrophoresis method.Results Adriamycin inhibited the proliferation of SW480 cells,and the inhibitory effects increased in a concentration-and time-dependent manner.Adriamycin decreased the telomerase activity of SW480 cells.Adriamycin made the structure of telomere oligonucleotide sequences repeat more than four times with GGG,form the compact secondary structure,so as to speed up the migration velocity.Conclusion Adriamycin could inhibit the proliferation of SW480 cells and decrease the telomerase activity of SW480 cells,which may be associated with the formation of guanine-quadruplexes induced by adriamycin.
