摘　　要：目的探讨胃癌组织miR-374b的表达及其临床意义。方法采用RQ-PCR法检测20例胃癌患者癌组织(A组)及癌旁正常胃黏膜组织(B组)miR-374b的表达。用阳离子脂质体转染miR-374b仿制品及抑制剂至胃癌细胞株MGC-803和SGC-7901中,通过细胞计数试剂盒(CCK-8)和集落形成实验,检测miR-374b对胃癌细胞增殖能力的影响。结果 RQ-PCR检测显示,A组miR-374b表达量少于B组(P<0.05)。体外实验发现,转染miR-374b疑似物后,胃癌细胞株MGC-803和SGC-7901的增殖能力较对照组降低,MGC-803集落形成数减少(P<0.05)。miR-374b的低表达与胃癌患者的年龄、性别构成比、肿瘤部位、分化程度及TNM分期均无明显相关性。结论 miR-374b在胃癌组织中表达减少,miR-374b高表达可抑制胃癌细胞的增殖。Objective To investigate the expression and clinical significance of miR-374b in gastric cancer. Methods The expression of miR-374b was detected by RQ-PCR in 20 fresh gastric cancer samples（group A） and tumor-adjacent normal gastric tissues（group B）. The miR-374b mimic and inhibitor were transfected, respectively, into two gastric cell lines of MGC-803 and SGC-7901 by cationic lipofectamine. Cell counting kit-8 （CCK-8） analysis was performed to evaluate the effect of miR-374b on cell proliferation. The colony formation tests were used to evaluate the cell proliferation ability of MGC-803 transfected with miR-374b mimic. Results RQ-PCR analysis showed that eighteen samples in group A showed lower expression of miR-374b than that in group B（P〈0.05）. There was no statistical correlation between the low expression of miR-374b and clinical pathological characteristics, such as age, gender, TNM staging and tumor differentiation of gastric cancer （P〈0. 05）. CCK-8 analysis showed that the proliferation of MGC-803 and SGC-7901, transfected with miR-374b mimic, was significantly decreased after transfection （P〈0. 05 ）i Concomitantly, the proliferation ability of MCdE-803 was inhibited by miR-374b mimic（P〈0. 05）. Conclusion miR-374b expression is down-regulated in gastric carcinoma. Low expression of miR-374b is not associated with clinical pathological characteristics of gastric cancer. Up-regulation of miR-374b in gastric carcinoma could inhibit the proliferation of gastric cancer cells.
