摘  要:目的:探索金属硫蛋白水解酶17(ADAM17)在胚胎发育过程中过表达与血管形成的关系.方法:鸡胚正常培养到第3天(E3 d),采用鸡胚带壳开窗培养方法,取出3~4 ml蛋清,密封开口继续培养至E5d,将pCAGGS-ADAM17和pCAGGS-GFP质粒通过活体电转基因导入视顶盖,电转后继续培养到胚胎发育的E12 d,取材、4%多聚甲醛固定、OCT包埋、切片后采用原位杂交检测ADAM17表达水平,免疫荧光组织化学分析ADAM17过表达对微血管形成的影响.结果:鸡胚发育过程,采用活体电转基因实现了ADAM17在视顶盖中的过表达,ADAM17过表达对微血管的形成具有一定的影响.结论:ADAM17过表达促进微血管数量的增加.Objective:To explore the relationship between ADAM17 overexpression with angiogenesis during chicken embryo development.Methods:Fresh eggs were cultured in normal condition for three days (E3),3-4 ml egg white was removed from egg,and then the open hole was sealed for continuing culture to E5.pCAGGS-ADAM17 and pCAGGS-GFP plasmid mix were injected into the optic tectum and then handled by in vivo electroporation technology.Tissues were obtained until E12,fixed with 4% PFA,embedded with OCT,and cut into frozen sections.In situ hybridization method was used to analyze ADAM17 overexpression and fluorescence irnmunohistochemistry method was used to analyze the relationship between ADAM17 overexpression with angiogenesis.Results:ADAM17 overexpression was achieved in the optic tectum during chicken embryo development,and affected the formation of microvessels.Conclusion:ADAM17 overexpression can promote the number increase of microvessels.
