摘 要:目的:观察少突胶质细胞转录因子2(O1ig2)过表达对小鼠胚胎干细胞(mESCs)向少突胶质祖细胞(OPCs)分化的影响.方法:将体外培养的mESCs分为空载体感染和Olig2-GFP慢病毒感染组,免疫荧光显色检测Olig2的表达.感染后9、14、21 d和28 d神经元-胶质细胞抗原2(NG2)免疫荧光显色观察mESCs向OPCs的分化情况.结果:Olig2-GFP慢病毒感染组mESCs经筛选后均为Olig2+/GFP+,而空载体感染组mESCs无Olig2表达.感染后9d,两组细胞均无NG2表达;感染后14d,Olig2-GFP慢病毒感染组NG2高表达,空载体感染组仅有少数细胞为NG2弱阳性;感染后21d与14 d相比,两组均无明显变化;感染后28d,两组细胞均有较强的NG2表达,组间无差异.结论:过表达Olig2能够加速mESCs分化为NG2+的OPCs样细胞.Objective:To observe the effect of transcription factor Olig2 overexpression on the differentiation of mouse embryonic stem cells (mESCs) into oligodendrocyte progenitor cells (OPCs).Methods:The mESCs were assigned to empty vector lentiviral infected mESCs (vector group) and Olig2-GFP lentiviral infected mESCs (Olig2 group) to test the expression of Olig2 by immunofluorescence staining.On the 9 d,14 d,21 d and 28 d after infection,the expression of neuron-glial antigen 2 (NG2) was detected using immunofluorescence staining to show the differentiation of mESCs into OPCs.Results:mESCs of the Olig2 group were all Olig2+/GFP+ cells after selection,while mESCs of the vector group did not express Olig2.On 9 d after infection,mESCs of neither group expressed NG2.On 14 d,mESCs in Olig2 group expressed a high level of NG2,while there were only fewer NG2+ cells in the vector group,which had no changes on 21 d.Until 28 d after infection,mESCs of both groups were all NG2+ cells without difference between each other.Conclusion:Olig2 overexpression can facilitate the differentiation of mESCs into NG2+ OPC like cells.
