桂皮醛对人乳腺癌细胞株MDA-MB-435S侵袭能力的抑制作用及与miR-27a表达的关系探讨  被引量:7

摘  要:目的探讨桂皮醛对人乳腺癌细胞株MDA-MB-435S侵袭能力的抑制作用及与其调节MDAMB-435S细胞miR-27a表达的相关性。方法聚碳酸酯膜细胞培养小室(transwell小室)法检测桂皮醛对MDA-MB-435S细胞侵袭能力的影响;脂质体2000转染miR-27a模拟物/抑制物联合实时定量聚合酶链式反应(reaI time-poIymerase chain reaction,Real-time PCR)及transwelI小室模型法探讨miR-27a表达在MDA-MB-435S细胞侵袭中的作用和桂皮醛对MDA-MB-435S细胞miR-27a表达的干预及其与影响细胞侵袭能力的关系。结果桂皮醛作用MDA-MB-435S细胞12 h后,穿过transwell小室的细胞数量均较对照组明显减少(P<0.05);miR-27a模拟物与miR-27a抑制物转染后MDA-MB-435S细胞miR-27a的表达量分别是对照组的962.07倍和40%,经miR-27a抑制物转染后MDA-MB-435S细胞穿过transwell小室的细胞数量显著下降(P<0.05);桂皮醛作用MDA-MB-435S细胞12 h后可下调其miR-27a的表达量(2^(-△△Ct)分别为0.56、0.18和0.18);先经miR-27a模拟物转染预处理后经桂皮醛作用的MDA-MB-435S细胞穿过小室的数量均较未预处理的对照组明显增多(P<0.05)。结论桂皮醛对人乳腺癌MDA-MB-435S细胞株的侵袭能力具有抑制作用;miR-27a的过表达在MDA-MB-435S细胞的侵袭能力中发挥重要作用;桂皮醛抑制MDA-MB-435S细胞的侵袭能力与下调其miR-27a的表达有关。Objective To explore the inhibitory effect of cinnamaldehyde on invasion capacities of human breast cancer cell line MDA-MB-435S and its relation with regulating the expression of miR-27a. Methods The effect of cinnamaldehyde on invasive capacities of MDA-MB-435S was measured by Tran- swell matrigel invasion assay. The effect of miR-27a expression on invasive capabilities of MDA-MB- 435S, the intervention of cinnamaldehyde in the miR-27a expression, and its relation with its effect on in- vasive capabilities were defected with liposome 2000 transinfection miRNA27a mimics/inhibitors, real time-polymerase chain reaction (Real-time PCR), and Transwell chamber model. Results Compared with the control group, the number of cells passing through the transwell chamber was more significantly reduced after treated by cinnamaldehyde for 12 h (P 〈0.05). The miR-27a expression was 962.07 times and 40% of that of the control group after transinfected by miR-27a mimics and miR-27a inhibitors. After transinfected by miR-27a inhibitors, the number of cells passing through the transwell chamber was more significantly reduced (P 〈0.05). The miR-27a expression of MDA-MB-435S was down-regulated by 12-htreatment of cinnamaldehyde (2^-△△Ct =0.56, 0.18, 0.18, respectively). The number of miR-27a mimics transinfection pretreated MDA-MB-435S cells passing through the transwell chamber increased more ob- viously than the number of un-pretreated MDA-MB-435S cells in the control group (P 〈 0.05). Conclu- sions Cinnamaldehyde could inhibit invasive capabilities of human breast cancer cell line MDA-MB- 435S. The over-expression of miR-27a played an important role in the invasive capability of MDA-MB- 435S. The inhibition of cinnamaldehyde on invasive capabilities of MDA-MB-435S cells was correlated with down-regulating the expression of miR-27a.
