摘  要:目的观察新生大鼠缺氧缺血性脑损伤(HIBD)脑组织Bax基因表达变化及亚低温干预对其表达的影响,探讨亚低温治疗发挥脑保护作用的可能机制。方法将新生7日龄SD大鼠120只采用Rice法建立新生大鼠HIBD模型,随机分成3组:假手术组,缺氧缺血组,亚低温干预组,实验组根据处死时间的不同又分为5个亚组:2h组,6h组,12h组,24h组,48h组,每组各8只。应用实时荧光定量聚合酶链反应(RT-PCR)在mRNA水平检测新生大鼠HIBD时Bax基因表达变化。结果 Bax mRNA在假手术组表达甚微,无明显变化;在缺氧缺血组2h即出现上升,12h达到高峰,12h^48h维持在高峰(P<0.05);亚低温干预组各时间点Bax mRNA的表达水平较缺氧缺血组明显降低(P<0.05)。结论 Bax mRNA参与了新生大鼠HIBD,亚低温干预可降低新生大鼠缺氧缺血性脑组织Bax mRNA的表达,从而发挥其可能的脑保护作用。Objective To observe the changes of Bax gene expression in brain tissue after hypoxia ischemia brain damage (HIBD),and explore the possible protection mechanism of mild hypothermia.Methods A total of 88 seven day old neonatal rat models of HIBD established by Rice method were randomly divided into three groups:Sham operated group,HIBD group,and mild hypothermia intervention group.The experimental group were further divided into five sub groups (n=8 each)based on different time points after HIBD (2 h,6 h,12 h,24 h,48 h),respectively.Fluorescence quantitative PCR (PT PCR)technique was used to de-termine the expression changes of Bax mRNA in brain tissues.Results There was no significant change in Bax mRNA expression in control group.The expression was increased at 2h after HIBD,12h to the peak level,12 48h gradually increased (P<0.05).Bax mRNA expression in mild hypothermia intervention group was significantly lower than that of HIBD group at each time points (P<0.05).Conclusion Bax mRNA participated in HIBD of neonatal rats.Mild hypothermia intervention could reduce the expression of Bax mRNA in brain tissue after HIBD.Therefore,hypothermia intervention had some neuroprotective effects on the brain tissues of neonatal rats of HIBD.
