摘 要:目的:观察流体静压力对骨髓间充质干细胞增殖、细胞骨架及成骨分化的影响,及RhoA发挥的作用。方法:分离培养BMSCs,随机分为对照组、RhoA激动剂组、拮抗剂组、压力组、激动剂+压力组及拮抗剂+压力组。流式细胞仪、共聚焦显微镜检测细胞周期、细胞骨架,Real-time PCR检测成骨标志基因。结果:压力依赖RhoA的活性促进细胞周期启动、细胞骨架装配及成骨分化早期标志基因表达。结论:RhoA在压力导致的细胞周期启动、骨架装配及成骨早期分化的过程中具调控作用。Objective: To observe the effects of hydrostatic pressure on proliferation, cytoskeleton and osteogenetic differentiation of bone marrow mesenehymal stem cells (BMSCs), and explore the role of RhoA in this process. Methods: BMSCs were isolated and cultured in vitro. After that the ceils were randomly divided into control group, RhoA agonist group , RhoA antagonist group, hydrostatic pressure group, hydrostatic pressure combine agonist group and hydrostatic pressure combine antagonist group. Cell cycle and cytoskeleton were examined by flow-cytometric and confocal microscopy. Osteogenic markers were evaluated by real-time PCR. Results: Hydrostatic pressure could promote the start of the cell cycle and cytoskeleton assembly, the process relied on the activity of RhoA. With the hydrostatic pressure loading regime, the mRNA expression of Col.I and OCN increased significantly in early stage of osteogenic differentiation by up regulate the activity of RhoA. Conclusion: RhoA plays an important role in the start of the BMSCs cell cycle, cytoskeleton assembly and early stage of osteogenie differentiation under the mechanical pressure.
