摘 要:目的:观察实验性牙齿移动过程中,大鼠牙周组织中磷酸化Janus激酶(JAK2)与信号转导因子和转录活化因子(STAT3)的表达和分布。方法:选用24只7周龄雌性Wistar大鼠,每只大鼠上颌左右两侧分别为实验侧和对照侧,建立大鼠实验性牙移动的模型。将大鼠随机分成3组,分别在加力后3、7、14d处死动物。制备标本,用免疫组化染色法检测p-Jak2和p-Stat3在大鼠牙周组织中的表达,并采用计算机图像分析软件Image J对各组p-Jak2和p-Stat3在牙周组织中的表达强度进行定量分析。结果:p-Jak2和p-Stat3在正常牙周膜中均有表达,且在受力3、7、14d后,p-Jak2实验组受压力侧和受牵张力侧均较对照组表达升高;p-Stat3在受力3、7d后,实验组受压力侧和受牵张力侧较对照组表达升高,但受力14d后,实验组受压力侧和受牵张力侧与对照组的表达并无明显统计学差异。结论:正常牙周组织中存在p-Jak2和p-Stat3,且p-Jak2和p-Stat3参与了牙周组织改建的过程,这说明Jak2/Stat3信号转导通路参与了牙周组织改建中细胞内的信号传递。Objective: To investigate the expression and distribution of phosphorylated Janus kinase 2(p-JAK2) and phosphorylated signal transducer and activator of transcription 3 (p-STAT3) in periodontal tissues ,and analyze the role of p-JAK2 and p-STAT3 in periodontal bone remodeling induced by experimental tooth movement. Methods: The model of experimental tooth movement was established in twenty-four female Wistar rats, which were 7 weeks old. The left side of jaws of each rat was set as experimental side , and the right side as control side. The modal rats were randomly divided into three groups and then were sacrificed after 3,7,14 days respectively. The upper jaw and periodontal tissue were selected and prepared. The expression of p-JAK2 and p-STAT3 was detected with S-P immunohistochemieal staining in the control group and groups treated for 3,7,14 days and computer image analyses was used to study the expression of p-JAK2 and p-STAT3 in the periodontal tissues of the rats. Results: The expression of p-JAK2 and p-STAT3 in the periodontal tissues of the control group was weak. And the expression of p-JAK2 was statistically higher on both tension and compression side in the experimental groups on 3th, 7th and 14th day. The experiment of p-Stat3 was higher on both tension and compression side in the experimental groups on 3th and 7th day, but there was no statistics difference between experimental side and control side on 14th day. Conclusion: p-JAK2 and p-STAT3 express in the periodontal tissue, and JAK2/STAT3 signaling pathway might participate and play a role in the remodeling of periodontium.
