摘  要:目的探讨β淀粉样蛋白(Aβ)对三磷腺苷(ATP)引起的大鼠海马神经元损伤的影响及其相互作用。方法孕18 d大鼠取其胎鼠迅速断头取出海马神经组织进行原代培养,实验分为正常对照组、ATP组、Aβ组、ATP+Aβ组、Suramin组。使用台盼蓝计数检测细胞存活率,全细胞电压钳记录细胞膜电流情况,钙成像系统检测细胞内Ca2+水平。结果 ATP组、Aβ组、ATP+Aβ组细胞存活率与正常对照组比较差异均有统计学意义(P<0.05);ATP+Aβ组和Suramin组细胞存活率比较差异有统计学意义(P<0.05)。ATP+Aβ组和无Ca2+灌流液组全细胞电流与ATP组比较差异有统计学意义(P<0.05);Suramin组与ATP+Aβ组全细胞电流比较差异有统计学意义(P<0.05)。与加药前比较,ATP组F340/F380比值明显增加(P<0.05);与ATP组比较,ATP+Aβ组F340/F380比值增加(P<0.05);与ATP+Aβ组比较,Suramin组F340/F380比值明显下降(P<0.05)。结论 Aβ可能通过激活ATP受体通道引起显著的钙超载,从而导致更严重的细胞损伤。Objective To explore the effect of β-amyloid protein(AI3) on hippocampal neuronal damage induced by adenosine triphosphate(ATP) of rats and their interaction. Methods The fetal rats were taken from rats which gestated for 18 days and then were decollated to take the hippocampus. The hippocampus was proceeded primary culture, and then the study were divided into normal control group, ATP group, Aβ group, ATP + Aβ group and Suramin group. Trypan blue staining was used to count the cell survival rate;whole-cell voltage-clamp was used to record membrane currents and calcium imaging system was used to detect the concentration of intracellular Ca^2+. Results There were statistic differences of cell survival rates between ATP,Aβ,ATP + Aβ group and control group(P 〈0.05) ;there were statistic differences of cell survival rates between ATP + Aβ group and Suramin group ( P 〈 0.05 ). There were statistic differences of whole-cell currents between ATP + Aβ group, without Ca^2+ peffusate group and ATP group( P 〈 0.05 ) ;there were statistic differences of whole-cell currents between Suramin group and ATP + Aβ group ( P 〈 0.05 ). Compared with before giving drug, the F340/Faso ratio of ATP group increased ( P 〈 0.05 ) ; compared with ATP group, the F340/F380 ratio of ATP + Aβ group increased ( P 〈 0.05 ) ; compared with ATP + Aβ group, the F340/F380 ratio in Suramin group decreased (P 〈 0.05 ). Conclusion Aβ may cause significant calcium overload by activating the ATP receptor channel and induce the more severe cell damage.
