摘  要:目的研究人脑动静脉畸形中富含半胱氨酸酸性分泌蛋白(Sparc)及Vcan基因的表达。方法收集手术切除的脑动静脉畸形患者7例,患者术前均无栓塞或放疗史,以4例顽固性癫痫患者行颞前脑叶切除术后显微分离的血管组织样本作为对照,提取标本中的总RNA,反转录为cDNA,应用实时荧光定量RT-PCR方法比较Sparc及Vcan基因在人脑动静脉畸形和对照血管中的表达。结果Sparc基因在脑动静脉畸形血管中的相对表达量是1.633,对照组相对表达量是1.000。Vcan基因在脑动静脉畸形血管中的相对表达量是1.627,对照组相对表达量是1.030。Objective To study the gene Sparc and Vcan expressed in human brain arteriovenous malformation ( AVM) . Method 7 samples taken from AVM patients receiving surgical therapy and 4 control brain vessel samples taken from temporal lobectomy for treating medically intractable seizure were collected. None of the AVM patients had received any neurointerventional therapy or radiotherapy before the operations. Total RNA was isolated from the samples and reversely transcribed into cDNA. Real time RT-PCR was used to analyse the Sparc and Vcan gene ex-pressed in AVM and control group. Result The relative expression of gene Sparc in AVM group was 1. 633 in con-trast to 1. 000 in the control group. And the relative expression of gene Vcan in AVM group was 1. 627 in contrast to 1. 000 in control group. Conclusion The up-regulated expression of gene Sparc and Vcan was confirmed and they might be involved in the angiogenesis and progression of brain AVM.
