四君子汤对结肠腺癌荷瘤小鼠癌性恶病质的影响

摘　　要：目的研究四君子汤对结肠腺癌荷瘤小鼠癌性恶病质的影响及可能作用机制。方法 30只Balb/c小鼠随机分为正常组、模型组、四君子汤组,每组10只。模型组和四君子汤组予右侧前背部皮下注射结肠腺癌CT26细胞约1×107个,皮下种植肿瘤细胞后第1天开始模型组和正常组灌胃0.2ml生理盐水,四君子汤组灌胃0.2ml四君子汤药液,相当于3.01g/(kg·d)药粉,各组均灌胃15天。记录不同时间小鼠体重、饮食总量、平均每天饮食量,16天后取材比较脏器重量、瘤重及生化指标、炎症因子白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)含量。结果与正常组比较,模型组小鼠在第5天开始饮食摄入总量较正常组与四君子汤组减少,在第11-16天平均进食量亦明显低于四君子汤组及正常组(P<0.05);四君子汤组小鼠体重第16天时较正常组低,但明显高于模型组(P<0.05);模型组与四君子汤组小鼠脏器重量、瘤重、生化指标比较差异无统计学意义(P>0.05);与模型组比较,四君子汤组血清TNF-α、IL-6明显降低(P<0.05)。结论四君子汤能有效减轻结肠腺癌荷瘤小鼠癌性恶病质症状,提高体重,抑制炎症因子TNF-α、IL-6可能其作用机制之一。Objective To study the effect of Sijunzi Decoction（ 四君子汤） on cancer cachexia in colon adenocarcinoma-bearing mice and its possible mechanism.Methods Thirty Balb /c mice were randomized into the normal group,model group and Sijunzi Decoction group,with 10 in each.The model group and Sijunzi Decoction group were subcutaneously injected with 1 × 107CT26 cells.The model group and normal group were gavaged with 0.2 ml of normal saline.The Sijunzi Decoction group was gavaged with 0.2 ml of Sijunzi Decoction（ equivalent to 3.01 g /kg·d powder）.The intragastric administration lasted for 15 days.The weight,total food intake and average daily food intake of mice were recorded.The mice were sacrificed 16 days later.The organ weight,tumor weight as well as the levels of interleukin-6（ IL-6） and tumor necrosis factor-α（ TNF-α were compared.Results Comparing with the normal group and Sijunzi Decoction group,the total food intake was decreased in the model group on the 5th day and was significantly decreased from the 11 th to 16 th day（ P〈0.05）.The weight of mice in Sijunzi Decoction group was significantly lower than that in the normal group but higher than that in the model group（ P〈0.05）.There was no significant difference between the model group and Sijunzi Decoction group in the organ weight,tumor weight and biochemical indicators（ P〉0.05）.Comparing with the model group,the levels of TNF-α and IL-6 were significantly decreased in Sijunzi Decoction group（ P〈0.05）.Conclusion Sijunzi Decoction can effectively relieve the symptoms and increase weight of colon adenocarcinoma-bearing mice,which may be relevant to inhibiting TNF-α and IL-6.
