摘 要:目的:比较Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ不同fimA基因型牙龈卟啉单胞菌(Porphyromonas gingivalis,P.gingivalis)对人脐静脉血管内皮细胞(HUVEC)的黏附和侵入能力。方法:将Ⅰ~Ⅴ基因型P.gingivalis菌悬液分别与贴壁培养的HUVEC共同孵育后,分别经细胞裂解、接种培养、菌落计数检测各型P.gingivalis对HUVEC的黏附率;并分别经抗生素处理、细胞裂解、接种培养、菌落计数检测各型P.gingivalis对HUVEC的侵入率。结果:Ⅰ~ⅤfimA基因型P.gingivalis菌株均能黏附并侵入HUVEC,其中以ⅡfimA基因型P.gingivali的黏附率、侵入率最高,与其他各型相比差异均有统计学意义(P〈0.05)。结论:ⅡfimA基因型P.gingivalis对人血管内皮细胞的黏附、侵入能力明显强于Ⅰ、Ⅲ、Ⅳ、Ⅴ各fimA基因型。AIM: To compare the adhesive and invasive capabilities of Porphyromonas gingivalis ( P. gin- givalis ) with type Ⅰ,Ⅱ,Ⅲ,Ⅳ and Ⅴ fimA genes to human umbilical vein endothelial Cells (HUVECs) . METHODS : Bacterial suspension of P. gingivalis with type Ⅰ,Ⅱ,Ⅲ,Ⅳ and Ⅴ fimA genes were respectively incubated and cultured with adhere-wall cultured HUVECs. The adhesive rates were obtained after cell lysis, inoculation and colony counting and then compared. The invasive rates were obtained after incubation with antibiotics, cell lysis, inoculation and colony counting and then compared. RESULTS : The differences of adhesive rates and invasive rates among type Ⅰ,Ⅱ,Ⅲ,Ⅳ and Ⅴ fimA genes of P. gingivalis were statistically different ( P 〈 0.05 ) . The adhesive and invasive rates of P. gingivalis with type Ⅱ fimA gene were the higherst ( P 〈 0.05 ) . CONCLUSION : The adhesive and invasive abilities of P. gingivalis with type Ⅱ fimA gene to human vascular endothelial cells are stronger than P. gingivalis with other types of fimA genes .
