摘 要:目的:观察恶性肿瘤模型小鼠和抑郁模型小鼠行为学差异,以及血府逐瘀口服液对其的影响,并探究其作用机理。方法:将雄性昆明小鼠随机分为7组,正常对照组8只,抑郁模型组、抑郁血府逐瘀组、抑郁黛力新组、肿瘤模型组、肿瘤血府逐瘀组、肿瘤黛力新组,每组各7只。抑郁小鼠模型采用慢性轻度不可预见性的应激法建立。肿瘤造模选用腹水型S180肉瘤小鼠皮下接种。两血府逐瘀组和两黛力新组小鼠分别给予血府逐瘀口服液、黛力新片灌胃干预治疗21 d。观察治疗前后小鼠行为学改变,治疗后5-HT含量,肿瘤小鼠的瘤重。结果:经过21 d的干预,与正常对照组及抑郁两灌胃组相比,抑郁模型组及肿瘤模型组小鼠糖水消耗量明显减少(P<0.01),垂直运动及水平运动活动度明显下降,且血府逐瘀组稍优于黛力新组(P<0.05);与肿瘤模型组相比,肿瘤血府逐瘀组、肿瘤黛力新组糖水消耗量明显增加(P<0.01);两种干预药物作用相似。在5-HT方面,正常对照组、抑郁血府逐瘀组含量较高,与其它各组比较差异具有统计学意义(P<0.05)。肿瘤血府逐瘀组瘤体较肿瘤模型组及肿瘤黛力新组明显缩小(P<0.01)。结论:应激及肿瘤可以导致实验小鼠出现行为学改变和5-HT含量降低的抑郁表现;血府逐瘀口服液能有效干预行为学改变和5-HT含量的降低,并能抑制肿瘤生长,作用优于黛力新片。Objective: To observe the behavioral differences betwen depression model mice and tumor model in mice and the effects of Xuefu Zhuyu Oral Liquid ( XZOL ) on the brain behavior and monoamine neurotransmitter 5-HT. To explore its mechanism. Methods: Male KM rats were randomly divided into the control group, which had 8, the tumor model group, the depression model group, the XZOL group with tumor, and the Deanxit group with tumor, the XZOL group with depression, and the Deanxit group with depression,7 in each group. The depressive rat model was established by chronic unpredictable mild stress method. We chose ascites S180 sarcoma in mice subcutaneous inoculation for tumor model. XZOL was administered to rats in the XZOL group by gastrogavage, while Deanxit was given to those in the Deanxit Tablet group by gastrogavage. The intervention lasted for two weeks. The behavioral changes were observed by sucrose water consumption test and open-field test for behavioral differences. The 5-HT contents were detected using ELISA. And then we compared the tumor weight of tumor mice. Resuh : After chronic stress stimulus, compared with normal control group and depression two lavage group, the depression group and model group mice tumor sucrose water consumption decreased significantly (P〈0.01)and the behavioral score significantly decreased and XZOL group was slightly better than Deanxit group ( P〈O.05 ). Compared with the tumor model group, the tumor therapeutic group, sucrose water consumption increased significantly ( P〈0.01 ). Two kinds of drug intervention are similar. For the 5-HT, normal control group had higher levels than XZOL group ( P〈0.05 ). Tumors in XZOL group were relatively smaller than tumor and tumor Deanxit group ( P〈0.01 ). Conclusions : Stress and tumor can lead to behavioral changes and lower 5-HT content of rats. The intervention of XZOL could fight against depression-induced behavioral changes and increase 5-HT content and inhibit tumor arowth, which is better than
