摘  要:目的探讨大鼠成肌细胞中是否表达音猬蛋白(sonic hedgehog,Shh)信号通路成分、Patched蛋白1(Ptc1)及胶质瘤相关癌基因蛋白1(glioma-associated oncogene homolog 1(Gli1)。方法取大鼠大腿骨骼肌,机械分离加混合消化液处理分离单细胞,体外培养及克隆纯化,原代培养大鼠成肌细胞。采用RT-PCR法检测第3代成肌细胞中Shh、Ptc1及Gli1 mRNA的表达。采用免疫荧光细胞化学染色及Western blotting法检测成肌细胞中Shh、Ptc1及Gli1的蛋白表达。结果通过RT-PCR方法检测到大鼠成肌细胞表达Shh、Ptc1及Gli1的mRNA,通过免疫细胞化学技术及Western blotting法检测到大鼠成肌细胞表达Shh、Ptc1及Gli1蛋白,表达部位为细胞浆。结论 Shh信号通路存在于大鼠成肌细胞中。[Objective] To investigate the expression of Shh, Ptcl and Glil which are the components of sonic hedgehog signaling pathway in myoblasts of rat. [Methods] The myoblasts were isolated with mixed digestive juice from muscle samples of rats, cultured and purified clonally in vitro. The expression of Sonic hedgehog protein (Shh), Patched 1 (Ptcl) and Glioma-associated oncogene homolog 1 (Glil) was detected in the 3rd passage cells by RT-PCR, western blotting and immunofluorescence cytochemistry. [Results] The gene expression of Shh, Ptcl and Glil was found in rat myoblasts by RT-PCR. These cells were immunostained positive for Shh, Ptcl, and Glil in the cytoplasmin, respectively. [Conclusion] The results show that sonic hedgehog signaling pathway exists in rat myobIats.
