摘 要:目的观察咪多吡对帕金森病模型大鼠黑质炎性因子肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β和干扰素(IFN)-γ表达的影响。方法 96只SD大鼠随机分为对照组(Sham组)、帕金森病模型组(PD组)、左旋多巴治疗组(Levodopa组)和咪多吡治疗组(Eldepryl组),各组随机分为模型制备成功后4 d组和8 d组2个亚组。采用颈背部皮下注射鱼藤酮制备帕金森病大鼠模型,分别用免疫组化法和Western blot法检测TNF-α、IL-1β和IFN-γ的表达。结果与对照组比较,模型组各时间点大鼠黑质中TNF-α、IL-1β和IFN-γ表达显著增多(P均<0.01),8 d组高于4 d组(P均>0.05);与模型组比较,咪多吡治疗组和左旋多巴治疗组各时间点黑质中TNF-α、IL-1β和IFN-γ表达显著减少(P<0.05或P<0.01),8 d组低于4 d组,有显著性差异(P均<0.05);与左旋多巴治疗组相比,咪多吡治疗组各时间点大鼠黑质TNF-α、IL-1β和IFN-γ表达明显减少(P<0.05或P<0.01),8 d组低于4 d组(P均<0.05)。结论咪多吡可通过抑制黑质炎性因子TNF-α、IL-1β和IFN-γ的表达,发挥对帕金森病的治疗作用。Inflammatory factors TNF- α, IL-1β and IFN- γ play an important role in the pathogenesis of Parkinson's disease. In this study, we aim to explore the effects of Eldepryl on expression of inflammatory factors TNF-α, IL-1β and IFN-γ in substantia nigra of Parkinson's disease model rats. Firstly, a total of 96 SD rats were randomly divided into sham group, Parkinson's disease model group, Levodopa treatment group and Eldepryl treatment group. Each group was divided into 4 days and 8 days subgroups after the success of model preparation.Parkinson's disease rat model were established by subcutaneous injecting rotenone. Then, the expressions of TNF-α, IL-1β and IFN-γ in substantia nigra were detected by immunohistochemistry and Western blot. We found that the expression of TNF-α, IL-1β and IFN-γ in substantia nigra and striatum were increased in the Parkinson's disease model group compared with that in the sham group(all P 0.01), and there were more expressions of TNF-α, IL-1β and IFN-γ at 8 d compared with those at 4 d(all P 0.05). The expressions of TNF-α, IL-1β and IFN-γin substantia nigra were reduced in the Eldepryl treatment group and the Levodopa treatment group compared with that in the Parkinson's disease model group(P 0.05, or P 0.01), and there were less expressions of TNF-α, IL-1βand IFN-γ at 8 d compared with those at 4 d(all P 0.05). Furthermore, the expressions of TNF-α, IL-1β and IFN-γ in substantia nigra were reduced in the Eldepryl treatment group compared with that in the Levodopa treatment group(P 0.05, or P 0.01), and there were less expressions of TNF-α, IL-1β and IFN-γ at 8 d compared with those at 4 d(all P 0.05). From these results we can concluded that inflammatory factors TNF- α,IL-1β and IFN- γ in substantia nigra of rats with Parkinson's disease are increased significantly, and Eldepryl could treat Parkinson's disease by reducing the expression of inflammatory factor TNF- α, IL-1β and IFN- γ in substantia nigra.
