摘 要:目的:探讨怀牛膝苯提取物的抗着床作用与子宫肥大细胞(mast cell,MC)的关系。方法:孕鼠自孕1d起连续灌眼怀牛膝苯提取物5d(2g/L×2ml/d×5d);取其孕 3~7d子宫做冰冻切片;用组织化学和免疫组织化学双重染色技术检测MC的数量、胞内TNF-α和表面ICAM-1的表达状况。结果:灌药后,孕3~7d子宫MC总数较对照组显著增多(P<0.001);INFL-α免疫反应阳性MC(TNF-α+MC)数也显著多于对照鼠(P<0.001),MC中的TNF-α阳性率(TNF-α+MC/MC总数)由对照组的约10%提高到30%左右;ICAM-1免疫反应阳性MC(ICAM-1+MC)较对照组也显著增加,但ICAM-1阳性率有不同程度下降。结论:怀牛膝可使孕鼠子宫MC数量显著增多、表达TNF-α的能力大幅度提高,这可能是怀牛膝抗着床的免疫学机制之一。Objective:To invetigate the relationship between anti-implantation of Achyranthes bidentata BI and mast cell(MC) in the uterus. Methods: The pregnant mice were treated with benzene extract of Achyranthes bidentata BI from pregnant 1 d(ig 2 g/L× 2 ml/d× 5 d) . The uterus of pregnant 3 - 7 d mice were used to cryostal sections. The doubling staining technique of histochemistry and immunohistochemistry was used to detect the number of MC and the expression of TNF-a and ICAM-1 in MC. Results: Achyranthes bidentata BI could increase apparently the total number of MC and the number of TNF-a positive and ICAM-1 postitive MC in the uterus of pregnant 3 ~ 7 d mice( P <0.001); And TNF-α positive rate also increased obviously but ICAM-1 positive rate decreased. Conclusion: Achyranthes bidentata BI can increase obviously the number of MC and the expression of TNF-α in MC. It could be one of anti-implantation mechanisms of Achyranthes bidentata BI.
