前列腺癌中细丝蛋白A表达的临床意义

摘　　要：[目的]探讨细丝蛋白A(filamin A,FLNA)在前列腺癌中的表达及意义。[方法]采用免疫组织化学方法、Western blot检测68例前列腺癌组织及距其癌组织边缘2cm以上的镜下未见癌浸润的正常组织中FLNA蛋白的表达情况。[结果]免疫组织化学结果表明,FLNA蛋白在前列腺癌组织的表达阳性率为38.2%,较正常前列腺组织(75.7%)明显降低(P<0.05)。Western blot结果表明,FLNA的蛋白在前列腺癌组织的相对表达量较正常前列腺组织明显降低,两者比较差异具有统计学意义(P<0.05)。FLNA蛋白表达与前列腺患者年龄、PSA水平无关(P>0.05);而与淋巴结转移、临床分期以及Gleason评分有关(P<0.05)。[结论]前列腺癌组织中FLNA蛋白表达明显减低,可能在前列腺癌发生的负反馈调节中起重要作用。[Purpose]To analyze lhe expression of filamin A（FLNA） in prostale cancer and its clinical significance. [Methods] FLNA protein expression in 68 cases with prostate cancer and in normal tissues without caneer infiltration which was adjacent to eancer tissue with distance 〉2cm,was detected by immunohistochemistry and Western blot methods. [Results] Immunohistochemistry, result siaowed that FLNA protein expression was significantly lower in prostate cancer tissue than that in normal tissues （38.2% vs 75.7%,P〈0.05）. Western blot result showed that the relative amount of FLNA protein in prostate cancer tissue was significantly lower than that in normal tis- sues（P〈0.05）. The level of FLNA protein expression was not correlated to age and PSA （P〉0.05）, but it was correlated to lymph node metastasis,clinical stage and Gleason score （P〈0.05）. [Conclusion] Low FLNA expression in prostate cancer might play an important role as a negative regulator in prostate carcinogenesis.
