摘  要:目的:研究白藜芦醇在白血病细胞株K562中对WT1表达的影响。方法:采用MTT法测定不同浓度白藜芦醇对K562细胞的增殖抑制作用,并计算IC50;根据IC50结果设立药物组、阴性对照组和阳性对照组,采用以RT-PCR和Western Blot法检测WT1 mRNA和蛋白表达情况。结果:1白藜芦醇抑制K562细胞的增殖,72h的IC50为97μmol/L;225μmol/L白藜芦醇可明显抑制K562细胞中WT1 mRNA和蛋白的表达。结论:白藜芦醇可显著下调K562细胞中WT1基因mRNA和蛋白的表达。Objective: To study the effects of resveratrol on the expression of WT1 in K562 cells. Methods: K562 cells were treated with different concentrations of resveratrol. The proliferation and inhibition effect of resveratrol in K562 cells was detected by MTT, and the IC50 was calculated. According to the results of ICs0, drug group, negative control group and positive control group were made, and the expressions of WT1 mRNA and protein were detected by RT-PCR and Western Blot methods, respectively. Results: (1)Resveratrol inhibited the proliferation of K562 ceils, and the IC50 was 97μmol/L at 72 hours. (2)The expressions of WT1 mRNA and protein in K562 cells were reduced with 25μmol/L resveratrol. Conclusion: The resveratrol could reduce the expressions of WT1 mRNA and protein in K562 cells.
