摘  要:目的探讨卵巢癌COC1细胞中大量表达p53正向凋亡调控因子(PUMA)在抑制COC1细胞中的作用。方法采用人端粒酶逆转录酶(hTERT)启动子调控的PUMA基因腺病毒载体(Ad-hTERT-PUMA)转染卵巢癌COC1细胞,通过细胞活性检测试剂盒和凋亡试剂盒研究COC1细胞的生长情况,同时分析表达PUMA的水平。结果转染Ad-hTERT-PUMA后,PUMA大量表达,抑制COC1细胞生长,促进细胞凋亡。结论 Ad-hTERT-PUMA成功转染卵巢癌COC1细胞并大量表达,起到了抑制卵巢癌细胞增长的作用。Objective To over-express the P53 upregulated modulator of apoptosis(PUMA) in ovarian cancer COC1 cells for playing the role to restrain the cell growth .Methods Adenovirus vector (Ad)-human telomerase reverse transcriptase promoter (hTERT)-PUMA was transfected into ovarian cancer COC1 cells .Then the growth of COC1 cells was studied by CCK-8 and the apoptosis kit and the level of PUMA was analyzed .Results After transfection of Ad-hTERT-PUMA ,PUMA was over-expressed in COC1 cells ,which restrained the cells growth and promoted the cell apoptosis .Conclusion Ad-hTERT-PUMA is successfully transfected into ovarian cancer COC1 cells and PUMA is over-expressed to play the role for restraining the cells growth .
