舒芬太尼对胃癌SGC-7901细胞活力的影响

摘　　要：目的通过不同浓度的舒芬太尼作用于胃癌SGC-7901细胞,研究舒芬太尼对胃癌细胞活力的影响。方法将对数生长期的胃癌细胞随机均分为对照组(C组)、舒芬太尼0.5nmol/L组(0.5nM组)、5nmol/L(5nM组)、50nmol/L(50nM组)和500nmol/L(500nM组)。分别以0、0.5、5、50、500nmol/L浓度的舒芬太尼作用于胃癌细胞,FDA/PI荧光双染观察染毒后的胃癌SGC-7901细胞存活情况;CCK-8试剂盒检测细胞活力;AnnexinV-FITC流式细胞术检测胃癌细胞的凋亡情况;最后运用细胞周期检测试剂盒分析胃癌细胞的细胞周期。结果 FDA/PI双染观察显示,随着舒芬太尼染毒浓度的增高,死亡的胃癌SGC-7901细胞比例不断增加,存活的正常细胞比例下降;CCK-8分析表明药物作用后的胃癌细胞的活力呈下降趋势,与C组比较,12h时500nM组和24、48h时50、500nM组细胞活力明显下降(P<0.05或P<0.01)。与C组比较,5nM组、50nM组和500nM组细胞凋亡率明显升高(P<0.05或P<0.01)。运用AnnexinV-FITC/PI凋亡实验发现胃癌细胞凋亡和死亡的数量也随药物浓度的增加而不断增多;最后通过检测染毒后的胃癌细胞,发现与C组比较,5nM、50nM和500nM组G2/M期细胞比例明显升高,S期细胞比例明显降低(P<0.01)。结论舒芬太尼可以使胃癌SGC-7901细胞的活力下降,促进胃癌细胞凋亡。Objective In this study,gastric cancer cell line SGC-7901 was exposed to different concentrations of sufentanil,in order to investigate the effects of sufentanil on SGC7901 cells.Methods Gastric cancer cells were divided into control group（group C）,sufentanil 0.5nmol/L group（group 0.5nM）,sufentanil 5nmol/L group（group 5nM）,sufentanil 50nmol/L group（group 50nM）,sufentanil 500nmol/L group（group 500nM）.Gastric cancer cells were exposed to sufentanil at the concentrations of 0,0.5,5,50,500nmol/L,respectively;FDA/PI staining was used to seperate living gastric cancer SGC7901 cells from dead cells;cell viability was analysed by CCK-8assay.Apoptosis of gastric cancer cells was analyzed by Annexin V-FITC/PI Flow Cytometry and cell cycle changes of gastric cancer cells were determined by cell cycle kits.Results As the concentration of sufentanil increased,the results from FDA/PI staining showed that the proportions of dead and apoptotic SGC-7901 cells increased.Cell vitality was observed decreased from CCK-8assay.Compared with gourp C,Cell vitality in group 500 nM at 12 hand group 50,500 nM at 24,48 hwere decreased significantly（P〈0.05 or P〈0.01）.The apoptosis rate in groups 5nM,50 nM and 500 nM were higher than group C（P〈0.05 or P〈0.01）.The foundings from Annexin V-FITC/PI Flow Cytometry displayed that apoptotic SGC-7901 cells increased with the concentration of sufentanil increased.And compared with group C,much more cells were arrested in G2/M phase in groups 5nM,50 nM and 500 nM while lower in S phase（P〈0.01）.Conclusion Sufentanil can inhibit the cell viability and promote the apoptosis of gastric cancer SGC-7901 cells.
