摘 要:目的以7-去氢胆固醇为底物,通过微生物转化合成25-OH-7去氢胆固醇。方法利用微生物转化法,结合TLC和HPLC分析,筛选出能够转化25-OH-7-去氢胆固醇的菌株,并对转化条件进行优化。结果从300株菌株中筛选出了5株能够转化25-OH-7-去氢胆固醇的菌株。转化产物经1HNMR及MS等鉴定,确认为25-OH-7-去氢胆固醇,用其中1株菌Pseudonocardia autotrophica CGMCC5098进行了转化条件的初步优化,转化率达25%。结论建立了一条制备25-OH-7-去氢胆固醇的新方法,为活性VD3的生产提供了一条新途径。OBJECTIVE To synthesize 25 - OH - 7 - dehydrocholesterol via biotransformation of microorganism using 7 - dehydro- cholesterol as substrate. METHODS The strains which had the capacity to convert 7 - dehydrocholesterol into 25 - OH - 7 - dehydro- cholesterol by biotransformation techniques with TLC and HPLC analysis were screened, and optimize the conversion conditions. RESULTS Five strains which had the capacity to convert 7 - dehydrocholesterol into 25 - OH - 7 - dehydrocholesterol were screened from three hundred of strains. The product was identified as 25 - OH - 7 - dehydrocholesterol on the basis of its 1HNMR and MS spec- tral data. The bioconversion ratio of 25 - OH - 7 - dehydrocholesterol reached 25% after optimization by using one of the five strains that was named Pseudonocardia autotrophica CGMCC5098. CONCLUSIONS This study creats a new method for the preparation of 25 - OH - 7 - dehydrocholesterol and provides a new way for producing the active vitamin D_3.
