摘 要:目的研究中医经验方剂牙周续补固齿汤对4种常见牙周炎致病菌的抑菌作用,明确药物汤剂中的有效抑菌成分和有效抑菌浓度。方法将药材通过煎煮、沉淀等程序,分别获得药物上清液与沉淀2种成分的粉剂,倍比稀释法分别配得10种浓度溶液。用钢管抑菌环法对4种牙周致病菌牙龈卟啉单胞菌、中间型普氏菌、巨核梭杆菌、粘性放线菌分别行抑菌实验,记录药液抑菌浓度。实验重复3次,取均值。统计分析获得2种药物成分对4种牙周致病菌的有效抑菌浓度。结果药物沉淀部分各浓度组对4种常见牙周致病菌的抑菌环均小于8mm。药物上清液6.25%以上浓度对牙龈卟啉单胞菌和中间型普氏菌的抑菌环大于12mm;12.5%以上浓度对粘性放线菌的抑菌环大于12mm;25.0%以上浓度对巨核梭杆菌的抑菌环大于12mm;药物上清液对4种牙周致病菌均有不同程度的抑菌作用。结论抑菌实验证实牙周续补固齿汤中有效成分为上清液。12.5%以上浓度对4种牙周致病菌均有不同程度的抑菌效果。上清药液抑菌效果由强到弱依次是牙龈卟啉单胞菌、中间型普氏菌、粘性放线菌和巨核梭杆菌。Objective To investigate the bacteriostatic effect of Yazhou Xubu Guchi decoction extract on four periodontal pathogens. Methods The herbs were prepared by boiling, precipitation and other procedures to obtain supernatant and precipitate two components of the drugs, and then air-dried to get two kinds of powder. Ten concentrations of the solution were prepared by coubling dilution and cultivated with four periodontal pathogens Porphyromonas gingivalis Aeromonas ( Pg ), PrevoteUa intermedia ( Pi ), Fusobacterium nuelcatum ( Fn ), Actinomyces viscosus ( Av ) under anaerobic conditions and 37~C for three days and repeated three times, respectively. The size of bacteriostatic ring was recorded. The results were statistically analyzed. Results All the bacteriostatic rings around all concentrations in precipitation group were less than 8mm. The bacteriostatic ring in supernatant group with concentration above 6. 25% was greater than 12mm for Pg and Pi and was greater than 12mm with concentration above 12. 5% for Av and above 25% for Fn. Conclusion The bacteriostatic experiments confirmed that the effective components of Yazhou Xubu Guchi decoction extract was the supernatant. The supernatant concentration above 12. 5% showed varying degrees of bacteriostatic effect on the four periodontal pathogens. The inhibitory effect from strong to weak was Aeromonas Porphyromonas gingivalis, Prevotella intermedia, Actinomyces viscosus and Fusobacterium nuelcatum.
