摘　　要：目的探讨干细胞转录因子Sox2对宫颈鳞癌细胞增殖能力的影响及其机制。方法将质粒pIRES-EGFP-Sox2转染宫颈鳞癌SiHa细胞系,采用G418筛选构建稳定表达Sox2的SiHa细胞克隆(SiHaSox2),MTT法检测Sox2表达对宫颈鳞癌细胞增殖能力的影响;流式细胞术检测细胞周期的变化;Western blot检测细胞周期相关蛋白CyclinD1表达变化。结果 pIRES-EGFP-Sox2转染SiHa细胞后(SiHa-Sox2组),细胞Sox2基因和蛋白均高表达(P均<0.01);SiHa-Sox2组细胞增殖速度加快;细胞周期中S期细胞比例明显增加;Western blot检测CyclinD1表达上调(P<0.05)。结论 Sox2基因通过上调CyclinD1蛋白的表达加速细胞周期进程,促进宫颈鳞癌细胞的增殖,从而促进宫颈鳞癌的发生发展。Objective To investigate the effect and its mechanism of stem cell related transcription factor Sox2 on the proliferation of cervical squamous carcinoma cell line SiHa.Methods Plasmid pIRES-EGFP-Sox2 or empty plasmid（pIRES-EGFP-empty）was stably transfected into SiHa cells.The expression of Sox2 was detected by both RT-PCT and Western blot.The effects of Sox2 on cellular proliferation and cell cycle were studied by MTT assay and flow cytometry（FCM）respectively.The expression of cell cycle related protein CyclinD1 was detected by Western blot.Results Compared to SiHa-EGFPcells,the expression of Sox2 was obviously up-regulated in SiHaSox2cells（P〈0.01）.MTT result showed that SiHa-Sox2 cells grew faster than the control cells.The overexpression of Sox2 increased the proportion of transfected cells in phase S.The increased expression of CyclinD1 was further detected after the successful expression of Sox2（P〈0.05）.Conclusion Sox2 could enhance the proliferation of cervical squamous cancer cells in the manner of up-regulating CyclinD1 expression.
