摘 要:目的:研究慢性不可预见性温和应激(CUMS)对大鼠的行为学和MOR1D表达的影响。方法:将12只大鼠采用随机数字表法分为对照组和CUMS抑郁模型组各6只。分别于应激前和21d应激后进行行为学观察,采用荧光定量PCR检测MOR1DmRNA的表达。结果:模型组糖水偏好试验显示糖水摄入减少,模型组旷场试验显示总行程、运动平均速度和直立次数减少,相比对照组均有显著性差异(P<0.001)。模型组伏隔核MOR1D mRNA表达较对照组高,有显著性差异(P<0.05)。结论:慢性应激后,大鼠出现的快感缺失改变与MOR1DmRNA表达下调有关。Objective:To investigate the changes of behavior and expression of mu opioid receptor1D(MOR1D)mRNA after chronic and unpredictable mild stress(CUMS).Methods:Twelve rats were randomly divided into control group(n=6)and model group(n=6).The rats in the model group were subjected to CUMS for 21 consecutive days.The behaviors of two groups were observed.The expression of MOR1 DmRNA was measured by real-time quantitative PCR.Results:The sugar consumption in the model group were significantly lower than those in the control group(P〈0.001).The total distance,speed and rearing were reduced in the model group,revealed by the open field test(P〈0.001).The expression of MOR1 DmRNA in the model group was significantly lower than that in the control group(P〈0.05).Conclusion:The down regulation of MOR1 DmRNA probably plays a key role in anhedonia of rats exposed to stress.
