摘  要:目的:观察上皮标志物和转录因子之间的相关性,从而确定这些因子在肿瘤上皮-间质转化(EMT)过程中所起的作用,进一步研究其与口腔鳞癌临床病理之间的关系,揭示其在口腔鳞癌发生发展中的作用及临床意义。方法:免疫组化SP法检测57例口腔鳞癌组织和10例正常口腔黏膜组织中E-cad、Snail、Slug、MMP-2、MMP-9表达,分析其与口腔鳞癌临床病理的关系及E-cad和Snail、Slug、MMP-2、MMP-9表达的相关性。结果:1.口腔鳞癌中E-cad、Snail、Slug、MMP-2、MMP-9的阳性表达率分别为29.82%、75.43%、82.46%、71.93%、78.95%。2.口腔鳞癌中E-cad的表达与分化程度有关(P<0.05),Snail、Slug、MMP-2、MMP-9的表达与分化程度无关(P>0.05);3口腔鳞癌中Ecad、Slug、MMP-2、MMP-9的表达与浸润和淋巴结转移有关(P<0.05),Snail与淋巴结转移和浸润无关(P>0.05)。4.口腔鳞癌中E-cad的表达与Snail、Slug、MMP-9表达均呈负相关(P<0.05),而与MMP-2的表达无关(P>0.05)。结论:转录因子与上皮标志物之间相关性明显,它们与口腔鳞癌的侵袭、淋巴结转移和分化程度相关。这些因子在EMT过程中不同阶段起作用,它们之间的相互作用是口腔鳞癌发生发展的重要环节。Objective: To study the relationship of epithelial markers and transcription factors and confirm their functions in OSCC EMT and investigate their relationship between these factors and clinicopathologic features of OSCC in order to further reveal their roles during OSCC progression. Method: SP immunohistochemical method was used to detect the expression of E-cad, Snail, Slug, MMP-2 and MMP-9 expression in 57 cases of OSCC and 10 cases of normal oral mucosa, analyze their relationship with clinicopathological factors of OSCC and correlation between E-cad and Snail, Slug, MMP-2, MMP-9 expression. Result: (1).In OSCC tissues,the positive rate of E-cad, Snail, Slug, MMP-2, MMP-9 respectively were 29.82 %, 75.43 %, 82.46 %, 71.93 %, 78.95 % .2 (1) Expression of E-cad was related to differentiation degree of OSCC (P 〈0.05), but Snail, Slug, MMP-2, MMP-9 were not (P〉0.05) : (2)Expression of E-cad, Slug, MMP-2, MMP-9 had significantly positively correlated with invasion and lymph node metastasis (P 〈0.05), Snail has nothing to do with the invasion and lymph node metastasis (P 〉0.05). 3 The expression between E-cad and Snail, Slug, MMP-9 expression was negatively correlated in OSCC (P 〈0.05), but not related to MMP-2 (P 〉0.05). Conclusion: Transcription factors and epithelial markers were correlated significantly, they were related to the invasion and lymph node metastasis and differentiation of oral squamous cell carcinoma. These factors play a role in the different stages of EMT process, the interaction of them is an important part of the development of OSCC.
