摘 要:目的通过检测14-3-3和Bcl-2蛋白在人脑胶质瘤中的表达情况,探讨其在胶质瘤发生、发展中的作用机制.方法采用免疫组化S-P法对5例正常脑组织标本和68例人脑胶质瘤标本进行14-3-3和Bcl-2蛋白检测.结果与正常脑组织比较,14-3-3蛋白在胶质瘤表达异常增强(P<0.05);在不同病理分级中胶质瘤组织表达无统计学意义(P>0.05),随着胶质瘤恶性程度的增加,14-3-3蛋白表达的强度和范围增加(P<0.05).Bcl-2蛋白在不同级别的恶性胶质瘤中阳性表达率有差别(P<0.05),且表达强度随着胶质瘤病理分级增高呈明显增高趋势.结论 14-3-3和Bcl-2蛋白对胶质瘤的恶性程度、肿瘤的病理分级有影响,可作为指导临床病理诊断及分级的参考指标.Objective To explore biological significance in the occurrence and development of glioma through investigating the expression of 14-3-3 and Bcl-2 proteins in human glioma. Methods The immunohistochemical S-P method was used to detect 14-3-3 and Bcl-2 proteins in 5 normal brain examples and 68 human glioma specimens. Results Compared with that in normal brain tissue,the expression of 14-3-3 protein was upregulated obviously( 〈 0. 05) in glioma tissues,but in different pathological grading glioma,there had no obvious difference(P〉0. 05),and the strength and scope of the 14-3-3 protein expression in glioma tissues increased with the increase in the malignant degree( P〈0. 05). The positive expression rate of Bcl-2 showed obvious differences in malignant glioma with different pathological grades(P〈0. 05),and the intensity of Bcl-2 expression presented a significant elevating trend with the increase in the pathological grade. Conclusion 14-3-3 and Bcl-2proteins in human glioma tissues can affect the malignant degree and pathological grade of glioma,so that they could be used as a guide for clinical pathological diagnosis and grading.
