摘 要:目的:研究5-羟基-3,7,3′,4′-四甲氧基黄酮的抗炎作用,并从影响一氧化氮生成探讨其机制。方法:采用二甲苯致小鼠耳廓肿胀法观察5-羟基-3,7,3′,4′-四甲氧基黄酮的抗炎作用;观察5-羟基-3,7,3′,4′-四甲氧基黄酮对LPS诱导的RAW264.7细胞释放NO的影响。结果:5-羟基-3,7,3′,4′-四甲氧基黄酮各剂量组均能明显减轻二甲苯所致的小鼠耳廓肿胀;在50μM时对LPS诱导的RAW264.7细胞释放NO有轻微的抑制作用。结论:5-羟基-3,7,3′,4′-四甲氧基黄酮具有抗炎作用,对抑制炎性因子一氧化氮的产生有轻微的抑制作用,可知其可能是通过其他机制产生抗炎作用的。Objective : To observe the anti-inflammatory effects of 5-hydroxy-3,7,3' , 4'-tetramethoxyflavone extracted from Callicarpa nudiflora and the possible mechanism from aspect of nitric oxide (NO) production. Methods: The anti-inflammatory effect of DHT-1 was observed in auricle edema mice models induced by xylene and influence of 5-hydroxy-3,7,3', 4r-tetramethoxyflavone on NO production in LPS-induced RAW264.7 cells were observed. Results: 5-hydroxy-3,7,3',4' tetramethoxyflavone at low-dose (0.05 g/kg), medium-dose (0.1 g/kg) and high-dose (0.2 g/kg) all could reduce the ear edema induced by xylene in mice. The NO production in LPS-induced RAW264.7 cells was slightly inhibited by 5-hydroxy-3,7,3' ,4'- tetramethoxyflavone at a concentration of 50/aM. Conclusions: 5-hydroxy-3,7,3',4'-tetramethoxyflavone has anti-inflammato- ry effect and a slight inhibition on NO production indicating it may exert anti-inflammatory effects through other mechanism.
