结肠癌患者血清中miRNA-21含量检测及其与VEGF水平的关系研究

摘　　要：目的:研究结肠癌患者血清中miRNA-21含量及其与血管内皮生长因子(VEGF)水平的关系。方法:选取在本院体检中心体检并确诊为结肠癌的120例患者纳入观察组,选择同期100例健康者纳入对照组,采用荧光定量PCR的方法检测miRNA-21的含量,采用酶联免疫吸附法检测VEGFA、VEGFB、VEGFC的含量,比较两组间的差异并分析miRNA-21与VEGF的关系。结果:观察组血清中miRNA-21的相对含量低于对照组,VEGFA、VEGFB、VEGFC的含量均高于对照组;血清中miRNA-21含量与VEGFA、VEGFB、VEGFC的含量呈负相关。结论:结肠癌患者血清中miRNA-21含量异常降低,且与血管新生因子VEGFA、VEGFB、VEGFC的含量相关,miRNA-21可能是调控结肠癌血管新生的潜在分子机制。Objective： To investigate serum content of miRNA-21 in colon cancer patients and its correlation with VEGF level. Methods： A total of 120 cases diagnosed as colon cancer in physical examination center of our hospital were enrolled as observation group, 100 cases of healthy persons in the same period were enrolled as control group. Then miRNA-21 content were detected by fluorescence quantitative PCR, VEGFA, VEGFB, VEGFC content were assayed by enzyme linked immunosorbent assay. Then the difference between two groups were compared and correlation between miRNA-21 and VEGF were analyzed. Results： Serum miRNA-21 content of observation group were lower than that of control group, but VEGFA,VEGFB, VEGFC content were higher than those of control group; serum miRNA-21 content were negatively correlated with VEG- FA,VEGFB, VEGFC content. Conclusion： Serum content of miRNA-21 in colon cancer patients is abnormally decreased and its content is correlated with angiogenesis factor VEGFA, VEGFB, VEGFC; miRNA-21 may be a potential molecular mechanism that regulate angiogenesis in colon cancer.
