子宫肌瘤及子宫平滑肌中myocardin的表达及意义

摘　　要：目的:检测心肌素(myocardin)在子宫肌瘤及子宫平滑肌中的表达,探讨其与子宫肌瘤的关系,及在子宫平滑肌中的表达规律,从而进一步指导临床诊疗工作。方法:收集30例患者的子宫肌瘤和子宫平滑肌组织,并进行原代培养;采用western blot方法检测子宫肌瘤和子宫平滑肌组织中的myocardin的蛋白表达情况,同时采用real-time PCR方法检测相应的mRNA表达情况。结果:myocardin在子宫肌瘤中的表达较子宫平滑肌中明显降低。在雌激素E2浓度梯度下,子宫平滑肌myocardin在1 nmol/L E2作用下表达明显增加,并且E2的这种促进作用能够被其抑制剂所减弱。结论:myocardin对子宫肌瘤的发生可能存在抑制作用,E2可进一步促进子宫平滑肌中myocardin的表达。Objective：To investigate the expression of myocardin in uterine smooth muscle and uterine leiomyoma. Methods：30 cases of uterine leiomyoma and paired uterine smooth muscle sample were collected and cultured. The expressions of myocardin protein and mRNA were examined by western blotting and real-time PCR. Results ：The expression of myocardin in uterine leiomyoma was significantly lower than uterine smooth muscle. E2 treatment significantly increased the expression of myocardin at the dose of 1 nmol/L,and this effect could be inhibited by E2 antagonist. Conclusions：Myocardin may suppress the occurrence of uterine leiomyoma and E2 can promote the expression of myocardin in uterine smooth muscle.
