摘  要:目的研究表没食子儿茶素没食子酸酯(EGCG)对食管癌细胞Eca109端粒酶活性及细胞凋亡的影响,探讨EGCG对食管癌细胞生长的抑制作用及其机制。方法 TRAP-PCR银染法检测30例不同食管组织标本(正常组织、癌旁组织、癌组织)的端粒酶活性。用不同浓度(0、25、50、100、200、400mg/L)EGCG作用于Eca109细胞,24、48、72h后采用MTT方法检测细胞增殖抑制率。用不同浓度(0、100、200、300mg/L)EGCG作用于Eca109细胞,24、48、72h后采用流式细胞仪检测细胞凋亡率,并采用TRAP-PCR银染法检测细胞端粒酶活性。结果 TRAP-PCR银染法检测显示,食管癌组端粒酶阳性率(86.67%,26/30)明显高于癌旁组(13.34%,4/30)和正常食管组(0.00%,0/30,P<0.05)。MTT检测结果显示,不同浓度EGCG作用于Eca109细胞后均表现出细胞生长抑制作用,且具有时间和剂量依赖性。EGCG作用于Eca109细胞后可导致细胞凋亡率增加及端粒酶活性降低,且具有时间和剂量依赖性。结论 EGCG可抑制Eca109细胞端粒酶活性,诱导细胞凋亡,从而起到抑制食管癌细胞生长的作用。Objective To study the effect of epigallocatechin-3-gallate (EGCG) on the telomerase activity and cell apoptosis of Eca109 ceils, and to explore the inhibitory effect of EGCG on esophageal cancer cells and its related mechanism. Methods The telomerase activity was assessed in 30 esophageal tissue specimens (normal esophageal tissue, tissue adjacent to carcinoma and carcinoma tissue) by TRAP-PCR argentation method. The inhibitory rate of Eca 109 cells as determined by MTT after being treated with various concentrations of EGCG (0, 25, 50, 100, 200, 400 mg/L) for 24, 48 and 72 h, respectively. The cell apoptosis rate and the telomerase activity of Ecal09 cells after being treated with various concentrations of EGCG (0, 100, 200, 300 mg/L) for 24, 48 and 72 h, respectively, were detected by flow cytometry and TRAP-PCR argentation method. Results The results of TRAP-PCR argentation showed that the telomerase positive rate was significantly higher in esophageal cancer tissue (86.67%, 26/30) than in carcinoma adjacent tissue (13.34%, 4/30) and normal tissue (0). MTT results showed that EGCG inhibited the growth of Eca 109 ceils in a dose and time dependent manner. EGCG induced apoptosis of the Eca 109 cell and reduced the telomerase activity of Eca 109 cells in a dose and time dependent manner. Conclusion EGCG can reduce the telomerase activity and induce apoptosis of Eca 109 cell, thus inhibit the growth of esophageal cancer cells.
