高效液相色谱-DAD法同时测定饲料中6种防腐剂

摘　　要：本实验建立了同时分离检测饲料中苯甲酸（BA）、山梨酸（SA）、对羟基苯甲酸甲酯（MP）、对羟基苯甲酸乙酯（EP）、对羟基苯甲酸丙酯（PP）和对羟基苯甲酸丁酯（BP）6种防腐剂的高效液相色谱-二极管阵列检测器（DAD）法。以甲醇和0.1 mmoL/L甲酸铵溶液（pH5.5）为流动相，结合溶剂梯度和流速梯度，在220～300 nm为提取最大吸收波长的条件下对这6种防腐剂进行分离与检测。结果表明，6种防腐剂在6 min内实现了基线分离，线性相关系数在0.9896-1.0000，BA和BP的检出限分别为0.1μg/mL和0.025μg/mL，SA、MP、EP和PP的检出限均为0.05μg/mL。样品加标平均回收率为85.5%～95.2%，相对标准偏差（RSD）为1.47%～3.25%（n=5）。A high performance liquid chromatography （HPLC）-diode array detector （DVD） method was developed for the determination of 6 kinds of preservatives including benzoic acid（BA）,sorbic acid（SA）,methylparaben（MP）,ethyl p-hydroxybenzoate（EP）,propylparaben（PP）and butylparaben（BP）with multi-gradients.Under velocity gradient combined with solvent gradient within 220 -300 nm with extracted maximum absorption wavelength,the six preservatives were sep-arated and detected using methanol and 0.1 mmoL/L ammonium formate solution （pH=5.5）as mobile phase.The results showed that complete sparation of the 6 preservatives was achieved within 6 min,the linear correlation coefficients were between 0.9896 and 1.0000,the detection limits were 0.1 μg/mL for BA,0.025 μg/mL for BP,0.05 μg/mL for BA,SA, MP,EP and PP,the average spiked recoveries were between 85.5% and 95.2%,and the relative standard deviation（RSD） was in the range of 1.47%~3.25%（n=5）.
