摘  要:目的:通过检测中度低温下肠上皮细胞屏障氧化应激损伤后炎症因子(TNF-α、IL-1β、IL-6)的表达水平,阐明中度低温对其炎症因子表达的影响及其临床意义。方法:应用Caco-2细胞株建立体外肠上皮细胞屏障模型,分中度低温组、常温组、亚低温组3个组,肠上皮细胞屏障氧化应激损伤后,应用逆转录PCR和实时荧光定量PCR检测细胞内炎症因子mRNA的表达水平,ELISA法检测培养上清中炎症因子的表达水平。结果:肠上皮细胞屏障氧化应激损伤后,中度低温组细胞内炎症因子的mRNA和以及培养上清中炎症因子的质量浓度明显低于常温组和亚低温组。结论:肠上皮细胞屏障氧化应激损伤后,中度低温比常温、亚低温更能抑制炎症因子的分泌,具有重要的临床意义。Objective:To illuminate the effect of moderate hypothermia on inflammatory cytokine expression and its clinical significance by detecting inflammatory cytokine(TNF-α、IL-1β、IL-6)expression levels after the intestinal epithelial cell barrier injured by oxidative stress. Methods:The Intestinal epithelial cell barrier model in vitro was established by Caco-2 cells,divided into moderate hypothermia group,normal temperature group and sub-hypothermia group. The expression levels of mRNA in the cells were examined by reverse transcription PCR and rea1 time PCR,and inflammatory cytokine in the cultural supernatant was measured by ELISA. Results:After the intestinal epithelial cell barrier injured by oxidative stress,the mRNA in inflammatory cytokines in moderate hypothermia group and the mass concentration of inflammatory cytokines in the cultural supernatant were significantly lower than those in normal temperature group and sub-hypothermia group. Conclusion:Moderate hypothermia can inhibit the secretion of inflammatory cytokine compared to normal tempera-ture and sub-hypothermia,which has important clinical significance.
