电针治疗对帕金森病大鼠认知功能的保护作用

摘　　要：目的研究电针治疗对帕金森病(Parkinson disease,PD)大鼠认知功能的影响。方法通过立体定位向大鼠脑部注射6-羟基多巴胺(6-OHDA)制备PD大鼠模型,筛选造模成功的大鼠进行为期两周的电针治疗与多巴丝肼药物治疗,Y迷宫评价大鼠的认知能力,通过分子生物学方法测算大鼠造模时受累的右侧半脑组织匀浆中乙酰胆碱转移酶(ChAT)的酶活性变化,免疫组化观察大鼠右侧半脑脑部海马区及纹状体部ChAT阳性纤维表达情况。结果与PD模型组大鼠相比,经电针灸治疗的大鼠学习记忆能力明显改善(P<0.05),脑部ChAT酶活性提高(P<0.01),光镜下可见海马区及纹状体区的ChAT阳性表达细胞数显著增加(P<0.01);电针灸治疗与多巴丝肼治疗组相比,大鼠的学习记忆能力及脑部ChAT表达无统计学差异(P>0.05)。结论电针灸治疗能明显改善PD大鼠认知功能,其机制可能与保护中枢乙酰胆碱神经元及提高ChAT表达有关。Objective To study the cognitive-improving effect of electrical acupuncture treatment on Parkinson＇ s disease （PD）-model rats.Methods The PD model was established by direct injection of 6-hydroxydopamine （6-OHDA） into the medial forebrain bundle using stereotactic technique.The successful PD models were treated with a two-week electrical acupuncture or Levodopa and Benserazide therapy,respectively.The choline acetyl transferase （ChAT） level in the right halves of the brain of the 6-OHDA-induced PD models treated with acupuncture or drugs were examined by tissue homogenization and immunohistochemical test （IHC）,and the cognitive function were assessed by Y-maze test.Results Compared with the PD model group,the study ability of the electrical acupuncture treatment group was improved （P＜0.05）,the brain ChAT activity was higher （P＜0.01）,and the ChAT-positive cell number in the hippocampus and striatum was increased （P＜ 0.01）.And there was no significant difference in study ability between acupuncture treatment and drug treatment （P＞0.05）.Conclusions Electrical acupuncture treatment improved cognitive function of PD models due to its potential protective role on acetylcholine neurons and up-regulating effect on brain ChAT level.
