摘 要:目的 探讨黄芪多糖对糖尿病大鼠血游离脂肪酸水平的影响及其机制.方法 选取SPF级雄性Wistar大鼠40只,完全随机分成4组,每组10只:空白对照组、糖尿病组[高热量饮食+小剂量链脲佐霉素(35 mg/kg)尾静脉注射后糖尿病造模成功继续高热量饮食]、黄芪多糖对照组[黄芪多糖400 mg/(kg·d)灌胃]和黄芪多糖干预组[糖尿病造模成功+高热量饮食+黄芪多糖400 mg/(kg·d)灌胃].给药5周后观察动物一般情况,检测血糖、血清游离脂肪酸和胰岛素水平.取肝脏组织,采用蛋白质印迹法测各组磷酸化腺苷酸活化蛋白激酶(p-AMPK)、总腺苷酸活化蛋白激酶(AMPK)、细胞膜脂肪酸转运酶/CD36(FAT/CD36)、肉毒碱棕榈酰转移酶(CPT1)蛋白表达水平.结果 糖尿病组动物血清游离脂肪酸、血糖水平及体质量明显高于空白对照组、黄芪多糖对照组与黄芪多糖干预组[分别为(0.54 ±0.13) mmol/L比(0.24±0.12)、(0.22 ±0.10)、(0.35±0.11) mmol/L,(20.5±3.2) mmol/L比(7.0±0.7)、(7.3±0.5)、(11.1±2.3)mmol/L,(223 ±3)g比(202 ±4)、(201 ±3)、(205 ±3)g,均P<0.05].黄芪多糖干预组血清游离脂肪酸和血糖水平低于糖尿病组(P<0.05),而各组胰岛素水平差异均无统计学意义(均P>0.05).糖尿病组动物肝脏p-AMPK、CPT1、细胞膜脂肪酸转运酶FAT/CD36蛋白表达明显低于其他3组(P<0.05).而各组总AMPK及FAT/CD36表达差异无统计学意义(P>0.05).结论 黄芪多糖可以通过非胰岛素途径活化AMPK降低糖尿病大鼠游离脂肪酸水平,改善胰岛素抵抗.Objective To investigate the effect of astragalus polysaccharides (APS) treatment on the level of blood free fatty acid and its mechanism.Methods Forty male Wistar rats were randomly divided into 4 groups:the control group,type 2 diabetes(T2DM) group [high-heat diet and a low dose of streptozotocin(35 mg/kg) by end intravenous injection],APS group [APS 400mg/(kg · d) by irritating the stomach with the solution] and APS and T2DM group [T2DM Wistar rats were treated with APS 400mg/(kg · d) by irritating the stomach with the solution].After treatment of 5 weeks,the levels of serum FFA and insulin were assayed;the expression of adenosine monophosphate activated protein kinase (AMPK),phosphorylated adenosine monophosphate activated protein kinase(p-AMPK),cytomembrane fatty acid translocase CD36 (FAT/CD36) and carnitine palmitoyltransferase I (CPT1) were assessed by Western blotting method.Results Serum free fatty acid (0.54 ± 0.13)mmol/L,blood glucose level (20.5 ± 3.2) mmol/L and body weight (223 ± 3) g of T2DM group were obviously higher than those of other groups (P 〈 0.05).Blood glucose levels (11.1 ± 2.3) mmol/L of Astragalus Polysaccharides and T2DM group were significantly lower compared with T2DM group (20.5 ± 3.2) mmol/L (P 〈 0.05),while insulin levels were not significantly different between T2DM group (P 〉 0.05).The levels of the p-AMPK,cytomembrane FAT/ CD36 and CPT1 expressions of Astragalus Polysaccharides were lower (P 〈 0.01),but the expression of total AMPK and FAT/CD36 showed no significant difference (P 〉 0.05).Conclusion APS can reduce the level of FFA in T2DM rat and improve the insulin resistance of liver tissue,and this protective mechanism may be relative to the AMPK activation.
