摘 要:目的观察针刺足三里、百会穴对阿尔茨海默氏病(AD)转基因快速老化SAMP8小鼠的学习记忆能力及海马区胰岛素受体(InsR)mRNA表达的影响,并探讨其作用机制。方法用Morris水迷宫进行筛选,将符合AD模型标准的SAMP8小鼠随机分为模型组、针刺治疗组(简称为针刺组),每组10只,并随机选取同源、同龄的10只SAMR1小鼠作为正常对照组。针刺组针刺小鼠足三里、百会穴,留针30 min,每日1次,10 d为1个疗程,疗程之间间隔2 d,连续治疗3个疗程。通过水迷宫隐蔽平台试验、空间探索试验判断小鼠的学习记忆能力,并通过逆转录聚合酶链反应(RT-PCR)检测左侧海马区InsR mRNA的相对表达量。结果隐蔽平台试验中,模型组平均逃避潜伏期(PLL)较正常对照组显著延长,有效区停留时间显著缩短(P<0.05);针刺组PLL较模型组显著缩短,有效区停留时间显著延长(P<0.05);平台停留次数3组间比较差异无统计意义(P>0.05)。空间探索试验中,模型组原平台象限百分比较正常对照组显著降低(P<0.01),跨越次数显著增高(P<0.05);针刺组原平台象限百分比较模型组显著升高(P<0.01),跨越次数显著减少(P<0.05)。模型组左侧海马区InsR mRNA相对表达量较正常对照组显著降低(P<0.05),针刺组较模型组显著升高(P<0.05)。结论针刺足三里、百会穴可通过上调海马区InsR mRNA的表达,从而提高AD SAMP8小鼠的学习记忆能力。Objective To conduct the experimental trial to observe acupuncture at "Zusanli"( ST36) and"Baihui"( GV20) on the improvement of learning and memory ability in transgenic rapid aging SAMP8 rats and the effect of hippocampus InsR mRNA expression. Methods Screen rats with Morris water maze first up-to-standard SAMP8 rats were randomized into model group( 10 cases) and acupuncture group( 10 cases). Another 10 homoage SAMR1 rats were control group. Needle at ST36 and GV20,30 minutes. The duration of treatment was 10times. 3 durations needed. Results Hidden platform trial: Average incubation period( PLL) of acupuncture group obviously shortened,compared model group,P〈 0. 05; and retention time of effective area prolonged,P〈 0. 05.Acupuncture group and control group without exception. Platform stops in three groups without significant difference. Space exploration trial: original platform quadrant percentage in acupuncture group heightened than model group,P〈 0. 01; and crossing frequency decreased,P〈 0. 05; no exception between acupuncture group and control group. PT-PCR tested InsR mRNA relative transcript level in left hippocampus in acupuncture group increased than model group,P〈 0. 05,without differences compared control group. Conclusion Acupuncture at ST36 and GV20 can raise hippocampus InsR mRNA expression to enhance SAMP8 rats learning and memory ability.
