摘 要:目的:探讨肝细胞肝癌(HCC)组织中miR-19的表达情况及其临床意义。方法:用qRT-PCR的方法检测51例HCC组织及其癌旁组织以及10例正常肝组织中miR-19的表达;分析了miR-19表达与HCC患者临床病理因素及预后的关系。结果:miR-19的表达量在正常肝组织、癌旁组织、HCC组织中依次增高,差异均有统计学意义(均P<0.05)。miR-19表达与HCC患者的肿瘤大小(P=0.029)、转移(P=0.011)、静脉侵犯(P=0.002)及AJCC分期(P=0.008)有关,而与年龄、性别、肿瘤分化、卫星灶、肿瘤数量及AFP无相关(均P>0.05)。miR-19高表达的患者5年生存率明显低于低表达的患者(P<0.05)。miR-19高表达以及转移、静脉侵犯均为影响HCC患者生存的独立风险因素(均P<0.05)。结论:miR-19在HCC组织中表达上调,并可能与HCC的增殖、转移有关;miR-19高表达提示预后不良。Objective: To investigate the miR-19 expression in hepatocellular carcinoma (HCC) tissue and its clinical significance. Methods: The miR-19 expression in 51 pairs of HCC tissues along with their matched adjacent tissues, and 10 samples of normal liver tissues were detected by qRT-PCR methods. The relations ofmiR-19 expression with the clinicopathologic factors and prognosis of HCC patients were analyzed. Results: The miR-19 expression level was increased in ascending order in normal liver tissue, tumor-adjacent tissue and HCC tissue, and the differences had statistical significance (all P〈0.05). The miR-19 expression was significantly related to tumor size (P=0.02), metastasis (P=0.011), venous invasion (P=0.002) and AJCC stage (P=0.008) of the HCC patients, but irrelevant to their age, sex, tumor differentiation, satellite lesions, tumor number and AFP level (all P〉0.05). The 5-year survival rate in patients with high miR-19 expression was significantly lower than that in those with low miR-19 expression (P〈0.05). High miR-19 expression togetherwith metastasis and venous invasion are risk factors affecting the survival of HCC patients (all P〈0.05). Conclusion: The miR-19 expression is up-regulated in HCC tissue, which probably may be associated with the proliferation and metastasis of HCC, and high miR-19 expression may predict an Unfavorable outcome.
