奥曲肽对人前列腺癌细胞株增殖与凋亡的影响

摘　　要：目的研究奥曲肽对雄激素非依赖性前列腺癌细胞DU145和雄激素依赖性前列腺癌细胞LNCaP增殖及凋亡的影响。方法对体外培养的人前列腺癌DU145和LNCaP细胞采用不同剂量奥曲肽(0.1、1、10、100和1000nmol/L)处理,采用XTT法测定细胞增殖,流式细胞法分析奥曲肽1000nmol/L对DU145和LNCaP细胞作用72h后细胞凋亡的影响。结果奥曲肽呈时间和剂量依赖性抑制DU145细胞的增殖,诱导其凋亡。奥曲肽1000nmol/L作用72h时,DU145细胞生长的抑制率达到57%。奥曲肽对LNCaP细胞生长的抑制和促凋亡作用不明显。结论奥曲肽对激素非依赖性前列腺癌有一定治疗效果,其作用可能通过直接地抑制细胞增殖和促进细胞凋亡。Objective To study the effects of octreotide on the proliferation and apoptosis of androgen-independent prostate cancer cell line DU145 and androgen-dependent prostate cancer cell line LNCaP. Methods The DU145 and LNCaP were treated with different concentrations of octreotide （0. 1,1,10,100 and 1000 nmol/L）. The proliferation of DU145 and LNCaP was detected .by XTT assay. The apoptosis of DU145 and LNCaP was analyzed by flow cytometry after the cell lines were treated with octreotide 1000 mnol/L for 72 hours. Results Octreotide inhibited the proliferation and induced the apoptosis of DU145 in the time- and dose-dependent manners. The inhibition rate of DU145 reached at 57% when it was treated with octreotide 1000 nmol/L for 72 hours. Octreotide had no significant effect on the proliferation and apoptosis of LNCaP. Conclusion Octreotide has a certain efficacy in treating androgen-independent prostate cancer, probably by directly inhibiting the proliferation and inducing the apoptosis of prostate cancer cells.
