摘 要:为筛选出具有较好除草活性的微生物并对其除草活性物质的发酵条件和分离纯化进行研究,本试验通过分离纯化带鱼体内微生物,测定代谢产物的除草活性,进而对影响代谢产物产生的各种因素进行研究,以明确其最适发酵条件,并对微生物所产生的除草活性物质进行分离纯化。结果表明,分离纯化得到的具有除草活性的菌株HY1、HY2、HY3均为细菌,HY2的除草活性较HY1和HY3强,其代谢产物对马唐的除草活性高于反枝苋。通过对HY2的发酵条件进行研究,发现该菌产生除草活性代谢产物的最适培养基为无机盐加糖培养基,最适发酵培养条件为起始pH值6.5、发酵温度25℃、摇瓶培养3-4d。培养滤液经液-液萃取、薄层层析分离和高效液相色谱分离后得到了一种保留时间为3.395min的除草活性成分。In order to screen the microbe with good herbicidal activity and research the fermentation conditions,isolation and purification of herbicidal active sunstance produced by the microbe,this experiment was studied on separating and purifing the microbes in hairtail gill and measuring the herbicidal active of metabolites,thus a variety of factors that affect metabolites produced was studied to find the optimum fermentation conditions.Also,the methods of isolation and purification of herbicidal active substance were established which laid the foundation for the structural identification.The results showed that the three microorganisms with herbicidal activity were bacteria,and the herbicidal activity of HY2 was stronger than HY1 and HY3.The metabolites got from HY2 had a stronger herbicidal activity on crabgrass than on redroot pigweed.According to the research of the fermentation conditions of HY2,the optimal medium for HY2 to produce metabolites with herbicidal active was inorganic salt medium with sugar and the optimum fermentation conditions were as follows:initial pH 6.5;fermentation temperature 25℃;incubation time 3-4d.An herbicidal active compound with the retention time of 3.395 min was obtained after the culture filtrate was separated by liquid-liquidextraction,TLC and HPLC.This study provides a new idea for exploring microbial herbicide,and it lays the foundation for the use of marine microbe and the development of new herbicides.
