摘 要:目的 研究神经纤毛蛋白-1(NRP-1)与CD4+ CD25+调节性T细胞(CD4+ CD25+Treg)在人脑胶质瘤组织中的表达及其临床意义.方法 收集人脑胶质瘤标本41例,其中Ⅰ级11例,Ⅱ级8例,Ⅲ级12例,Ⅳ级10例(胶质母细胞瘤).另取11例因颅脑创伤行内减压术患者的正常脑组织标本作为对照组.应用RT-PCR技术检测人脑胶质瘤组织和正常脑组织NRP-1的mRNA表达.流式细胞术检测上述组织中CD4+ CD25+ Treg细胞的比率.结果 NRP-1 mRNA在胶质瘤和正常脑组织中均有不同程度表达,但在胶质瘤中的表达量明显高于正常脑组织,且在胶质瘤中表达程度随其病理分级的增高而增高,差异有统计学意义(P<0.05);胶质瘤组织CD4+ CD25+ Treg细胞比率明显高于正常脑组织(P<0.01),在胶质瘤组织中CD4+ CD25+ Treg细胞的比率随肿瘤病理分级增高而增高,差异有统计学意义(P <0.05);NRP-1 mRNA表达程度和CD4+ CD25+ Treg细胞比率呈显著正相关(r=0.723,P<0.05).结论 NRP-1和CD4+ CD25+ Treg在脑胶质瘤中高表达,二者表达程度均与肿瘤的病理分级密切相关,NRP-1和CD4+ CD25+ Treg表达上调可能与肿瘤微环境的免疫耐受有关.Objective To investigate the expression and significance of Neuropilin-1 (NRP-1) mRNA and CD4 + CD25 + Treg in human glioma tissues.Methods The specimens of 41 patients with gliomas,including 11 cases of grade Ⅰ,12 grade Ⅱ,8 grade Ⅲ and 8 grade Ⅳ,were studied in the present study.11 cases of normal brain tissues from patients performed decompression operation resulting from cerebral hernia were selected as the control group.The expression of NRP-1 mRNA was investigated by reverse transcription polymerase chain reaction (RT-PCR) and the contents of CD4 + CD25 + Treg by flow cytometer in the glioma tissues and normal brain tissues.Results NRP-1 mRNA and CD4 + CD25 + Treg were expressed in different degrees in normal brain tissues and gliomas.The expression level in tumor tissues was higher than that in the normal brain tissues.The expression was related to the pathological grades of gliomas.The NRP-1 expression was significantly positively related with the CD4 + CD25 + Treg expression (P 〈 0.05).Conslusions The mRNA expression levels of NRP-1 and CD4 + CD25 + Treg were significantly higher in human gliomas which were positively associated with the grades of gliomas.NRP-1 and CD4 + CD25 + Treg might be related to the immunological tolerance in tumor microenvironment of human gliomas.
