摘  要:目的 探讨树突状细胞(DC)-细胞因子诱导的杀伤细胞(CIK)联合化疗对转移性大肠癌患者外周血微小RNA (miRNA)-29a表达的影响.方法 将39例转移性大肠癌患者随机分为两组,DC-CIK组23例采用XELOX方案+DC-CIK进行治疗,对照组16例采用XELOX方案进行治疗,采用实时荧光定量聚合酶链反应(FQ-PCR)法检测治疗前及3个周期后两组患者miRNA-29a表达的水平,并观察患者的疗效及评分.结果 经3个周期治疗后,DC-CIK组、对照组miRNA-29amRNA相对表达量均下降,分别为0.515±0.216比1.556±0.228;1.116 ±0.186比1.569±0.289,差异有统计学意义(P<0.05);与对照组比较,实验组下降更明显(P<0.05).处理组有效率(73.9%)明显高于对照组(31.3%,P <0.05).两组Ⅲ+Ⅳ度白细胞减少率的比较差异有统计学意义(P<0.05).Kamofsky评分比较,DC-CIK组提高优于对照组(P<0.05).结论 DC-CIK细胞可有效提高大肠癌整体治疗疗效.下调miRNA-29a是其重要机制之一.Objective To explore the effect and mechanism of dendritic cells (DC)-cytokine-induced killer cell (CIK) combined with chemotherapy on microRNA(miRNA)-29a mRNA in peripheral blood of metastic colorectal cancer.Methods Thirty nine patients with metastic colorectal cancer (CRC) were randomly divided into the treatment group (n =23,treated with DC-CIK combined with chemotherapy) and the control group(n =16,treated with chemotherapy).miRNA-29a mRNA level in peripheral blood of CRC patients was determined after 2 cycles by real-time fluorescent quantitative polymerase chain reaction (FQ-PCR).The efficacy and Karnofsky of each group were studied.Results The level of miRNA-29a mRNA of two groups was significantly decreased (P 〈 0.05),as compared with that before chemotherapy.The treatment group reduced the level of miRNA-29a mRNA more effective than the control group (P 〈0.05).Total efficacy of treatment group (73.9%) was significantly higher than that of control group (31.3%,P 〈0.05).There was significant difference in rate of hypolekocytosis at Ⅲ+ Ⅳ degree between two groups (P 〈 0.05).Treatment group improved better than the control group in the Karnofsky score (P 〈0.05).Conclusion DC-CIK can enhance the therapeutic effect probably by reducing the level of miRNA-29a mRNA.
