摘 要:目的 探讨新生期大鼠反复惊厥后海马区γ-氨基丁酸(GABA)A受体(GABAA R) α1和γ2亚单位表达的改变及其意义.方法 72只健康新生大鼠随机分为反复惊厥组(RS组)和对照组(CONT组),两组又随机分为10日龄、35日龄、70日龄亚组,各亚组12只大鼠.通过三氟乙醚吸入诱导建立新生期大鼠反复惊厥动物模型.采用Western blot法及逆转录-PCR法检测大鼠海马区GABAAR α1和γ2亚单位蛋白及mRNA的表达.结果 与CONT组比较,RS组大鼠10日龄亚组海马区GABAARγ2亚单位蛋白表达明显降低(P <0.01);35日龄亚组和70日龄亚组海马区GABAAR α1和γ2亚单位蛋白表达均明显降低(均P<0.05).与CONT组比较,RS组大鼠10日龄亚组、35日龄亚组和70日龄时亚组海马区GABAAR α1和γ2亚单位mRNA表达均明显降低(P <0.05~0.01).结论 新生期大鼠反复惊厥后海马区GABAAR α1和γ2亚单位表达明显降低,这可能在发育期反复惊厥所致的远期GABA介导的抑制功能下降中起重要作用.Objective To explore the changes and their significances of expressions of γ-aminobutyric acid (GABA) A receptor (GABAAR) α1 and γ2 subunits in hippocampus of rats after recurrent neonatal seizures.Methods Seventy-two healthy neonate rats were randomly divided into recurrent seizure group (RS group) and control group (CONT group).Both of the two groups were then divided into postnatal days 10,35 and 70 subgroups,with 12 rats in each subgroup.Recurrent neonatal seizure model was induced by inhalant flurothyl.Western blot and fluorescence quantitative reverse transcription-PCR were employed to determine the expressions of protein and mRNA of GABAAR subunits α1 and γ2 in hippocampus.Results Compared with CONT group,the protein expression of GABAAR subunit γ2 in hippocampus in RS group was significantly decreased at postnatal days 10 subgroup (P < 0.01) ; and the protein expressions of GABAAR α1 and γ2 subunits in hippocampus in RS group were significantly decreased at postnatal days 35 subgroup and days 70 subgroup (all P < 0.05).Compared with CONT group,the mRNA expressions of GABAAR α1 and γ2 subunits in hippocampus in RS group were significantly decreased at postnatal days 10 subgroup,days 35 subgroup and days 70 subgroup (P < 0.05-0.01).Conclusions The expressions of GABAAR oα1 and γ2 subunits in hippocampus are decreased significantly in rats after recurrent neonatal seizures.This phenomenon might play an important role in long-term GABA-mediated inhibition reduction induced by early life recurrent seizures.
