摘  要:采用高效液相色谱法同时测定明党参组培和栽培品中7-羟基香豆素、花椒毒酚、欧前胡素和珊瑚菜内酯4种香豆素成分的含量。Agilent-C18色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈—水溶液梯度洗脱,流速1.0 mL/min,柱温30℃;检测波长为314 nm,进样量为10μL。实验结果表明,栽培明党参的根皮4种香豆素成分7-羟基香豆素、花椒毒酚、欧前胡素、珊瑚菜内酯的含量普遍偏高,分别为5.8970、1.2710、0.0301、0.9236mg/g,明党参愈伤组织诱导根中7-羟基香豆素、花椒毒酚含量较高分别为3.7939、0.8384 mg/g;该方法简便、快速、具有良好的重现性,可用于明党参组培和栽培品中香豆素成分的含量测定。In this study,the concentrations of 7-hydroxycoumarin, xanthotoxol, imperatorin and phellopterin in the tissue culture and cultivated samples of Changium smyrnioldes were determined by HPLC. The Agilent-Cls column (250 mm x 4.6 ram,5 ttm) was used for the chromatographic separation with a mobile phase of acetonitrile-water solution in gradi- ent elution mode. The flow rate was 1.0 mL/min. The column oven temperature was controlled at 30 ℃. The detection wavelength was set at 314 nm and the injection volume was 10 p,L. The experimental results showed that the eoncemra- tions of the 4 investigated coumarins of the cultivated sample in cultivated Chang/urn root cortex was generally higher, which were 5. 8970,1. 2710,0.0301 mg/g and 0. 9236 mg/g, respectively. The concentrations of 7-hydroxycoumarin and xanthotoxol in callus induction of Changium root were 3. 7939 mg/g and 0. 8384 rag/g, respectively. The developed HPLC method was simple, accurate and reproducible. It was suitable for assaying the contents of comnarins in the tissue culture and the cultivated sample of C. smyrnioides.
