利用巴西蘑菇菌糠制备嗜热霉菌菌剂

摘  要:为了将巴西蘑菇菌糠作为嗜热霉菌菌剂的主要培养基质,比较了8株嗜热霉菌在巴西蘑菇菌糠培养基上的生长情况。结果表明,M2,M22,M28菌株在巴西蘑菇菌糠培养基上的羧甲基纤维素酶(CMC)酶活、过氧化物磷化酶(MnP)酶活较高,有效活菌数较多。培养基的初始水分含量和pH值对这3株嗜热霉菌菌剂的CMC酶活、MnP酶活和有效活菌数有较大的影响。通过比较小麦麸皮、小米粉和玉米粉3种辅料及其不同添加比例对3株嗜热霉菌的影响,确定了小麦麸皮为最适辅料。M2菌株在pH值为7、初始水分含量为24%、添加5%小麦麸皮的菌糠培养基上,M22菌株在pH值为9、初始水分含量为24%、添加10%~15%小麦麸皮的菌糠培养基上,M28菌株在pH值为9、初始水分含量为32%、添加10%小麦麸皮的菌糠培养基上的CMC酶活和MnP酶活最高,有效活菌数目最多,培养时间最短。最后,通过正交试验确定了M2,M22,M28菌株进行组合培养的最适培养条件为:pH值9,初始水分含量24%,小麦麸皮添加量15%,培养时间5 d。In order to utilize Agaricus blazei fungal chaff as the main substrate culturing thermophilic molds,the growth of 8 strains of thermophilic molds in Agaricus blazei fungal chaff medium was compared.The results showed that M2,M22 and M28 strains had higher CMC and MnP enzyme activities and more effective number of living molds in the fungal chaff medium.Different initial moisture contents and pH values of media could affect significantly the CMC and MnP enzyme activities as well as the number of living molds of M2,M22 and M28 strains.By comparing the effects of adding different radios of wheat bran,rice flour or corn flour into the media on the growth of 3 strains of thermophilic molds,wheat bran was proved to be the optimal auxiliary cultivation material.In order to attain the highest CMC and MnP enzyme activities and number of living molds as well as shortest culture time,M2 strain should be cultured in the medium of pH 7,the initial moisture content of 24%,adding 5% wheat bran;M22 strain should be cultured in the medium of pH 9,the initial moisture content of 24%,adding 10%-15% wheat bran;M28 strain should be cultured in the medium of pH 9,the initial moisture content of 32%,adding 10% wheat bran.Finally,the optimum culture conditions for the combined-culture of M2,M22 and M28 strains were attained by the orthogonal test.The strains should be cultured in the medium of pH 9,the initial moisture content of 24%,adding15% wheat bran and culturing 5 days.
